维生素E对人胚胎凋亡及体外发育的影响

摘    要:目的探讨抗氧化物维生素E(Vitamin E)对人类体外培养胚胎凋亡及体外发育的影响,并观察培养过程中胚胎凋亡的形态学变化。方法以常规体外受精-胚胎移植(in vitro fertilization and embryotransfer,IVF-ET)后废弃的胚胎为材料,培养过程中添加抗氧化物质维生素E,观察其对胚胎体外发育的影响,采用TUNEL方法对胚胎进行染色,观察其凋亡情况;用Hoechst33342对胚泡进行荧光染色,根据胚泡细胞总数及凋亡形态学变化对其凋亡进行检测。结果当添加100μmol维生素E时,与对照组相比,显著提高人类扩张期囊胚形成率和孵化率(P〈0.05),降低其凋亡率(P〈0.05)。结论维生素E能够降低人类囊胚的凋亡率,提高其体外发育率。

The effect of Vitamin E on the apoptosis and development of human embryos in vitro

Abstract:Objective To investigate the effect of antioxidant Vitamin E on human embryo apoptosis and development in vitro and to observe the morphology change of apoptosis embryo. Methods In the process of embryo culture in vitro, the embryos were stained with TUNEL, meanwhile the blastocysts were stained with Hoechst33342 fluorescence dye. According to total nuclei number of per blastocyst and the morphology changes, the situation of embryo apoptosis was examined. Results The rate of blastocyst expansion and the hatching rate were improved, at the same time the apoptosis rate was decreased in VitE group in which 100 μmol Vitamin E was added to the culturing medium that the difference were significant ( P 〈 0. 05 ) between two groups. Conclusion The antioxidant Vitamin E can reduce apoptosis rate and improve the development rate of human blastocyst in vitro.
