摘    要:目的采用HPLC法测定人血浆中盐酸安非他酮的质量浓度。方法采用Dikma DiamonsilC18色谱柱(250 mm×4.6 mm,5μm);柱温:30℃;流动相:甲醇-50 mmol.L-1NaH2PO4(含体积分数0.1%三乙胺和体积分数0.05%H3PO4)(体积比为55∶45);流量:1.0 mL.min-1;检测波长:248 nm;内标物为盐酸美西律。结果盐酸安非他酮线性范围为5.0~500μg.L-1。提取回收率均大于90%,日内、日间精密度均小于10%。结论该方法灵敏、准确,适用于临床血药浓度监测和药物动力学研究。

Abstract:Objective To establish a HPLC method for the determination of bupropion hydrochloride in human plasma.Methods Dikma Diamonsil C18 HPLC column(250 mm×4.6 mm,5 μm) was used;column temperature was 30 ℃;the mobile phase consisted of 50 mmol·L-1 sodium dihydrogen phosphate buffer(included 0.1% triethylamine and 0.05% phosphoric acid)-methanol(V∶V = 45∶55);the flow rate was 1.0 mL·min-1;the detection wavelength was 248 nm;mexiletine hydrochloride was used as internal standard.Results The linear range of bupropion hydrochloride was 5.0-500 μg·L-1.The extraction recovery of bupropion hydrochloride was more than 90%,intra-day RSD and inter-day RSD were less than 10%.Conclusions This method is proved to be sensitive,fast and accurate.Therefore,it can be used for therapeutic medicine monitor and pharmacokinetic study.
