溶性苯二氮受体的制备与分析

摘    要:采用新型去垢剂CHAPS制备兔大脑前叶皮质溶性苯二氮(艹卓)受体。溶膜受体放射配基结合分析,见苯二氮(艹卓)受体具有可饱和性,与3~H]FNP结合的亲和力KD_1为0.31nml/L,KD_2为6.7nmol/L。光亲和标记结合SDS电泳测得亚基分子量为55000;凝胶过滤层析测得受体分子量为632000。

Abstract:The benzodiazepine receptors in frontal lobes of cerebral cortex of rabbits were solu-bilized with the detergent 3- (3eholamidopropyl)-dimethylammonio propaneslfonate (CHAPS). 3H flunitrazepam binding to soluble receptors was saturable and showed nonlinear Scatchard plot with KD1 0.31 nmol/L and KD2 6.7 nmol/L. The molecular weight of the soluble benzodiazepine receptors determined by SDS-PAGE after photoaffinity labelling was 55 000, by chromatography on Sephadex G 200 was 623 000.
