兔VX2肝癌模型制作方法改进及影像学评价

摘    要:目的改进兔VX2肝癌模型的制作方法,同时评价其影像学特征。方法将日本大白兔60只分为开腹穿刺(A组)、开腹包埋(B组)、超声引导下18G针(C组)和16G针(D组)经皮穿刺四组(每组15只)。接种2周后,用超声、CT、MR检测肿瘤生长情况。结果A组、B组与D组比较原位种植率差异无统计学意义(P〉0.05),异位种植率有统计学意义(P〈0.05);C组与D组比较种植率有统计学意义(P〈0.05)。2周后超声检查肿瘤在声像图上表现为肝内低回声肿块,肿块周边及内部可见星点状、短条状血管分支。CT平扫肿瘤为略低密度病灶,增强扫描以肿瘤周边强化为著;MR平扫肿瘤呈稍长T1、长T2信号。结论超声引导下16G针穿刺制作的肿瘤模型方法简单、接种率高、肿瘤呈单个生长,其影像学表现类似人类原发性肝细胞肝癌,是适合介入治疗实验研究的动物模型。

Abstract:Objective To improve the creation of rabbit model bearing VX2 liver tumor, compare the imaging performance of ultrasound, CT and MRI in rabbit bearing VX2 liver tumor. Methods VX2 tumor was implanted in 60 Japan white rabbits by four methods: laparotomic puncture, laparotomic burying, 18G needle and 16G needle puncture under ultrasound guidance percutaneously, 15 rabbits per group. Two weeks after inoculating, tumor growth was monitored by ultrasound, CT and MRI. Results The success rate of original neoplasia was significantly higher in the group A, B and D compared with group C. The ectopic implantation rate was significantly higher in the group A and B compared with group D. Ultrasound showed that the tumor in the rabbit liver was spherical or sphere-alike hypoechoic nodule, and there were blood flow signals in and around the nodle. VX2 tumor in the liver appeared as a solitary low density nodle on unenhanced CT scanning. On MR scanning , the VX2 liver tumors were seen as hyperintensity on T1WI and slight hyperintensity on T2WI. Conclusion The establishment of model by 16G needle puncture under ultrasound guidance percutaneously was easier, success rate highly and single node. The image manifestations of the VX2 tumor in rabbits are similar to those of hepatic primary carcinoma in human.
