黄芪预处理大鼠心肌保护效应的实验研究

摘    要:目的 本文研究黄芪预处理对大鼠心肌的保护效应以及心肌保护效应与热休克蛋白之间的关系。方法 用在体大鼠复制缺血再灌注损伤模型,将大鼠随机分成对照组、缺血再灌组、黄芪组三组、观察心肌组织学、SOD酶的活性、MDA含量变化、心肌梗死范围和HSP70蛋白的表达的变化。结果 黄芪预处理组明显减少梗死面积和减少MDA的产生,保护细胞中SOD酶活性和超微结构,HSP70表达水平明显高于对照组及缺血再灌注组。结论 黄芪处理可减少缺血再灌注心肌损伤,并能诱导心肌细胞HSP70的表达。

Abstract:Objective To observe the effects of astragali(AS) pretreatment on myocardial ischemia reperfusion injury (IRI) in rat and to investigate the relations between protective effects of astragali and heat shock protein 70. Methods A model of IRI was produced by ligation left descending of coronary artery in Sprague Dowley. Rats were randomly divided into three groups: the control ( n =12);IRI group( n =24);Astragali treated group( n =24). The myocardium pathology were investigated by microscopy and electromicroscopy. MDA and SOD contents were determined as a marker of OFR(oxygen free radicals) injury.The levels of heat shock protein 70 were examined by immunohistochemical staining (LSAB methed). Infarct sizes also were measured. Results Astragali significantly reduced tissue MDA production ( P <0 01), myocardial infarct size ( P <0 01) and increased tissue SOD activity( P <0 01). HSP70 levels in astragali group were higher than those of IRI and the control group. Conclusions AS can prevent the ischemic injury of myocardium which are associated with the increases of expression of HSP 70 against myocardial IRI.
