摘 要: | 目的:探讨Ent-11α-hydroxy-15-oxo-kaur-16-en-19-oic-acid(5F)对人绒癌JAR细胞侵袭、转移、黏附的影响及MMP-9、VEGF表达的变化。方法:用不同浓度的5F作用JAR细胞,MTT比色法,Transwell小室,细胞黏附实验观察其增殖、侵袭、迁移、黏附能力的变化;Western blot方法检测5F对JAR细胞MMP-9、VEGF表达水平的影响。结果:作用6 h,5F能抑制JAR细胞的增殖、转移、侵袭、黏附,其抑制率呈剂量效应(P<0.05)。5F作用24 h JAR细胞,MMP-9、VEGF蛋白表达水平能明显下调。结论:5F能抑制JAR细胞的侵袭、转移、黏附并下调MMP-9、VEGF蛋白表达水平。

Abstract: | Objective:To explore the effect of Ent-11α-hydroxy-15-oxo-kaur-16-en-19-oic-acid(5F) on invasion,metastasis,and adhesion of human choriocarcinoma JAR cells and the changes of matrix metalloproteinase-9(MMP-9) and vascular endothelial growth factor(VEGF) expressions.Methods:JAR cells were treated with 5F of different concentrations.MTT colorimetric method,Transwell assay,and cell adhesion test were used to observe the changes of proliferative ability,invasive ability,metastasis,and adhesive ability of JAR cells;Western blot was used to detect the effect of 5F on the expression levels of MMP-9 and VEGF in JAR cells.Results:After treatment for 6 hours,5F could inhibit the proliferation,metastasis,invasion,and adhesion of JAR cells,the inhibitory rate showed a dose-dependent manner(P<0.05).5F could down-regulate the expression levels of MMP-9 and VEGF after treating JAR cells for 24 hours.Conclusion:5F can inhibit the invasion,metastasis,and adhesion of JAR cells,and it can down-regulate the expression levels of MMP-9 and VEGF. |
