脱落酸诱导Tca8113细胞株的动物实验研究

摘    要:目的: 检测不同剂量的植物激素脱落酸对人舌鳞癌Tca8113细胞株细胞凋亡的体内诱导。 方法: 通过人舌鳞癌Tca8113细胞株建立裸鼠移植瘤并随机分为对照组和实验组,实验组(0.5mmol/L和1mmol/L)局部肿瘤内直接注射脱落酸。检测其肿瘤生长抑制率,常规HE组织学观察和Caspase-3 mRNA原位杂交检测。 结果: 实验组移植瘤组织中癌细胞密度减少,细胞皱缩,胞浆红染,癌组织分化渐成熟,出现角化细胞,有角化珠形成趋势;间质结缔组织增多,淋巴细胞浸润。对照组瘤周组织中,有大量新生血管形成。0.5mmol/L和1mmol/L脱落酸对裸鼠移植瘤的抑制率分别为26.7%、36.1%,两者间存在显著差异(P<0.01)。实验组脱落酸作用后的瘤重与对照组间存在显著差异(P<0.001)。0.5mmol/L和1mmol/L脱落酸实验组经原位杂交检测Caspase-3 mRNA阳性表达位于胞浆呈棕黄色,部分细胞胞核也呈棕黄色,其阳性表达率分别为0.27±0.02、0.22±0.02,两者间有统计学意义(P<0.001);而对照组移植瘤的胞浆基本上不着色。实验组间与对照组间Caspase-3 mRNA的表达量也存在着统计学意义(P<0.01)。裸鼠的全身重要脏器(如肺、肾等)无器质性改变。 结论: 脱落酸不仅能抑制人舌癌Tca8113移植瘤的生长,而且还对移植瘤有分化诱导作用,导致Caspase-3的活性增加,促进细胞发生凋亡,为以后的临床应用研究提供了实验数据。

Abstract:Objective: To study the apoptosis of the Tca8113 squamous cell carcinoma cell line induced by different doses of abscisic acid (ABA) in animal models. Methods: A squamous cell carcinoma model was established in nude mice using the Tca8113 cell line. The model animals were randomly divided into the experimental group and the control group. ABA at two different concentrations (0.5mmol/L and 1mmol/L) was injected directly into tumors of mice in the experimental group. The inhibitory rate was observed and detected by staining with hematoxylin and eosin and in situ hybridization. Results: In the experimental group, the density of cancer cells was decreased. Horny pearls were seen in tumor tissues with some infiltrating lymphocytes in connective tissues. In the control group, a large number of new vessels were observed. In the experimental group, the tumor inhibitory rate of ABA was 26.7% at 0.5 mmol/L and 36.1% at 1mmol/L, with a signifi-cant difference between the 2 groups (P<0.01). A significant difference was found in the tumor weight between the experi-mental group and the control group (P<0.001). For the experimental group, in situ hybridization showed the presence of Caspase-3 mRNA in the cytoplasm and some cell nuclei. Expression of Caspase-3 mRNA was not observed in the control group. The rate of positive expression was 0.27± 0.02 in the tumors treated with ABA at 0.5 mmol/L and 0.22± 0.02 in those treated with ABA at 1mmol/L. No structural changes in the important organs such as the lungs, kidneys, etc. were observed in either group. Conclusion: ABA can suppress transplanted Tca8113 squamous cell carcinoma growth in nude mice by increasing the expression of Caspase-3 mRNA and inducing apoptosis.
