养血清脑颗粒对D-半乳糖致衰老大鼠慢性脑缺血模型脑组织中VEGF及HSP70的影响

摘    要:目的:观察养血清脑颗粒对D-半乳糖致衰老大鼠慢性脑缺血模型脑组织中VEGF、HSP70表达的影响,并探讨其作用机制。方法:60只Wistar大鼠随机分为正常组、模型组、尼莫地平组、养血清脑颗粒低剂量组(简称低剂量组)、养血清脑颗粒高剂量组(简称高剂量组)5组。在D-半乳糖(D-gal)致衰老的基础上永久性结扎大鼠双侧颈总动脉制备衰老大鼠慢性脑缺血模型。术前1d,高低剂量组分别用0.32g/mL、0.16g/mL养血清脑颗粒混悬液灌胃,尼莫地平组用尼莫地平混悬液灌胃,模型组灌服等量蒸馏水,正常组用等量生理盐水颈背部皮下注射。4W后,断头开颅取脑,处死大鼠,测定脑组织中VEGF及HSP70含量。结果:与正常组比较,模型组脑组织血管内皮中VEGF表达无明显改变,HSP70表达明显减少(P〈0.05)。与模型组比较,高剂量组、低剂量组脑组织血管内皮中VEGF、HSPT0表达增高(P〈0.01)。与低剂量组比较,高剂量组脑组织血管内皮中VEGF、HSP70表达增高(P〈0.01,P〈0.05)。结论:养血清脑颗粒使缺血脑组织中VEGF及HSP70的表达增加,提示该药可能从促进血管侧支循环形成及抑制神经元死亡等方面保护缺血脑细胞。

Abstract:Objective:To observe the effect of Yangxue Qingnao granule on the expressions of VEGF and HSP70 in brain of chronic cerebral ischemia models in D-galactose aging rats.Methods : Chronic cerebral ischemia models were induced by bilateral common carotid arteries ligation.60 rats were randomly divided into normal group,model group,nimodjpine group Yangxue Qingnao granule treatment group 1(low dosage)and group 2(high dosage ).The expressions of VEGF and HSP70 in brain of chronic cerebral ischemia models in D-galactose aging rats were measured by immunohistochemical method. Results :Compared with the model group,the expressions of VEGF and HSP70 were significantly increased in Yangxue Qingnao granule treatment group 1 and group 2.Conclusions:Yangxue Qingnao granule could protect cerebral ischemia injury through improving the formation of collateral circulation and inhibiting neuronal death.
