摘    要:目的:研究组蛋白去乙酰化酶抑制剂在体外和体内对胆囊癌细胞系和肝外胆管癌细胞系生长的影响,并探讨其临床应用价值。方法:用组蛋白去乙酰化酶抑制剂-曲古抑菌素A(trichostatin A,TSA)作用于胆囊癌细胞系(Mz-ChA-1)和肝外胆管癌细胞系(QBC939、KMBC、OZ),用MTT法检测TSA对这些细胞系生长的影响。将Mz-ChA-1接种裸鼠皮下建立胆囊癌裸鼠种植模型,观察TSA处理前后体内种植瘤生长的变化。结果:TSA可以抑制Mz-ChA-1和QBC939、KMBC、OZ的增殖,以上作用与药物浓度在一定范围内呈正相关。成功建立胆囊癌裸鼠体内种植模型,TSA处理后裸鼠体内种植瘤生长受抑制。结论:TSA在体外能抑制胆囊癌细胞系和肝外胆管癌细胞系的增殖;TSA在体内能抑制胆囊癌细胞系的增殖。

Abstract:Objective: To study the effects of inhibitor of histone deacetylase on the growth of gallbladder carcinoma cell line and extrahepatic cholangiocarcinoma cell lines in vivo and in vitro,and to investigate the perspective of histone deacetylase on clinical application. Methods: The survival rate of gallbladder carcinoma cell line(Mz-ChA-1 cell line) and extrahepatic cholangiocarcinoma cell lines(QBC939,KMBC and OZ cell lines) treated with different dose of TSA were detected using methylthiazoy tetrazolium(MTT) assay. The transplanted gallbladder carcinoma nude mice models were successfully established,the growth change of transplanted tumor treated with TSA was measured. Results: TSA can inhibit the proliferation of gallbladder carcinoma cell line(Mz-ChA-1 cell line) and extrahepatic cholangiocarcinoma cell lines(QBC939,KMBC and OZ cell lines). These effects were dose-dependent. The gallbladder carcinoma transplanted nude mice model was successfully established,and the growth of cancer was inhibited in the nude mice transplanted with gallbladder carcinoma cell line with TSA treatment. Conclusion: TSA can inhibit the growth of extrahepatic cholangiocarcinoma cell lines and gallbladder carcinoma cell line in vitro. TSA can inhibit the growth of gallbladder carcinoma cell line in vivo.
