摘    要:目的:建立一种高效液相色谱法测定胃内容物中安定含量的方法.方法:用外标法,以正常大鼠胃内容物作为空白对照组,以正常大鼠胃内容物添加安定标准品作为样品组,通过液-液萃取法提取样品、挥干并以流动相溶解后,用高效液相色谱仪进行定量分析,计算胃内容物中安定含量,并评价方法的准确度、精密度和线性范围.结果:胃内容物中安定的线性方程为Y=35.553 3X+40.383 3,r=0.999 5,线性范围为1.562 5~100 mg/L,最低检出限为0.076 mg/L,平均回收率为83.20%,日内精密度分别为4.89%、6.55%、7.22%,日间精密度为7.56%,均小于10%.结论:高效液相色谱法检测胃内容物中安定含量具有准确度和灵敏度高的优点.

Abstract:Objective: To establish a high performance liquid chromatography (HPLC)method for the analysis of diazepam in gastric contents. Methods: An external standard method was used. The stomach contents of normal rats were used as the control group, and those added diazepam standard compounds were used as the sample group. The samples were extracted by liquid-liquid extraction, evaporate, and dissolved with 200 p,1 mobile phase, and then were analyzed by HPLC. Diazepam contents in gastric contents were calculated. The accuracy, precision, and linear range of this method were evaluated. Results: The linear formula of diazepam in stomach contents was Y = 35. 553 3X + 40. 383 3 ,r = 0. 999 5, the linear rang was 1. 562 5 ~ 100 mg/L. The lowest detective limit was 0.076 mg/L, the average recovery rate was 83. 20%. The within-day relative standard deviation (RSD) were 4.89%, 6.55%, 7.22% ,respectively and the between-day RSD was 7.56%, and less than 10%. Conclusion: This method is precise and sensitive. It can be used for the diagnosis of diazepam poisoning.
