补肾化痰祛瘀方对血管性痴呆大鼠脑组织星形胶质细胞影响的实验研究

目的观察补肾化痰祛瘀方对血管性痴呆（VaD）大鼠脑组织星形胶质细胞的影响。方法采用改良线栓法制作大鼠VaD模型，缺血2h分别再灌注6h、24h、48h、72h、28d，随机分为正常组、假手术组、模型组、阿米三嗪组和补肾化痰祛瘀方组，并分别施以相应的处理。于不同再灌注时间点检测大鼠脑组织GFAP的表达。结果正常组与假手术组大鼠脑额顶叶皮层缺血半暗带区可见GFAP阳性细胞散在分布，假手术组与正常组阳性细胞数和平均光密度相近；模型组大鼠GFAP表达在6h增加，24、48、72h表达进一步增加，72h最高，28d仍维持较高水平，与正常组阳性细胞数和平均光密度差异显著；补肾化痰祛瘀方组于6、24、48、72h时可见GFAP的表达增加，与模型组阳性细胞数和平均光密度相近，于28d增加明显，与模型组差异显著；阿米三嗪组在不同时段可见GFAP的表达增加，与模型组阳性细胞数和平均光密度部分时段差异显著（72h、28d）；补肾化痰祛瘀方组与阿米三嗪组阳性细胞数和平均光密度在28d时差异显著。结论补肾化痰祛瘀方可调节脑缺血星形胶质细胞的GFAP表达，可能对损伤具有一定修复作用。

Experimental Study of Invigorating the Kidney Eliminating Phlegm Resolving Stasis Recipe on Effect of Astrocyte in Cerebral Tissue on Vascular Dementia in Rats

Objective:To study the effects of Invigorating the kidney eliminating phlegm resolving stasis recipe on astrocyte in cerebral tissue on vascular dementia in rats.Methods:The VaD rats were randomly divided into normal group, sham-operation group, model group, almitrine-raubasine group and invigorating the kidney eliminating phlegm stasis recipe group which undergo reperfusion at 6th, 24th, 48th, 72nd hours and 28th days after focal cerebral ischemia for 2 hours with the application of improved thread embolism. At different reperfusion time, the ability of behavioral memory of each group of rats was separately tested with step down test, expression of GFAP in brain was tested by immunohistochemistry SABC method.Results:GFAP positive cells scattered in the brain cortex ischemic penumbra of the normal group and sham-operation group rats. Compared with the normal group, sham-operation group had no obvious difference in positive cells amount and AOD.The GFAP expression of the model group's rats increased at the 6th hour, and further incerased at 24th, 48th and 72nd hour, among which the highest level was at the 72nd hour. There were obvious difference in positive cells amount and AOD compared with the normal group,and a high level was still maintained at the 28th day. GFAP expression was observed at different time in almitrine-raubasine group. Positive cells amount and AOD were different at different time periods from the model group(72h, 28day).Conclusion:Invigorating the kidney eliminating phlegm resolving stasis recipe may control GFAP expression of cerebral ischemia AS, and may modify injures to some degree. It is subject to a further study on its functional mechanism.
