维生素D_3体外羟化反应的实验研究

摘    要:本文利用肝脏羟化酶及肾脏羟化酶,对维生素D_3的体外25位羟化作用及1α位羟化作用进行了初步研究。结果表明:应用佝偻病鸡的肝线粒体及微粒体悬液,分别与维生素D_3、NADPH孵化系统在体外进行孵育,均可得到羟化产物—25-OH-D_3,线粒体维生素D_3-25羟化酶的活性为0.49Pmol 25-OH-D_3/2h·10mg pr,微粒体为0.28 pmol 25-OH-D_3/2h·10mg pr。二者转化率分别为11.9%与5.8%,线粒体25-羟化酶活性明显高于微粒体。应用佝偻病鸡的肾线粒体悬液,与25-OH-D_3、NADPH孵化系统在体外进行孵育,可以获得羟化产物-1a,25-(OH)_2-D_3,其转化率为37.5%。

Abstract:The 25-hydroxylation of vitamin D3 and 1α-hydroxylation of 25- (OH) -D3 in vitro were investigated. We used the rachitic chicken liver mitochondrial 25-hydroxylase system and microsomal 25-hydroxylase system to hydroxylate vitamin D3 in vitro. Mitochondrial 25-hydroxylase activeity was 0. 49 pmol 25-(OH) -D3/2h·10mg pr. Microsomal 25-hydroxylase activity was 0. 49 pmol 25- (OH) -D3/2h·10mg pr. Microsomal 25-hydroxylase actvity was 0. 28pmol 25- (OH) -D3/2h· 10mg pr. The transformation rate was 11.9% and 5. 8% respectively. The mitochondrial 25-hydroxylase activity was higher than the microsomal 25-hydorxylase.We also used the rachitic chicken kidney mitochondrial 25-hydroxyvitamin D3 1α-hydroxylase system to hydroxylate 25-hydroxyvitamin D3 in vitro. Its transformation rate was 37. 5%.
