| 摘 要： | 目的观察大鼠局灶性脑缺血再灌注后海马Bcl-2、Bax蛋白的表达及参附注射液对其的影响。方法成年雄性SD大鼠30只,随机分为假手术组、模型组、参附注射液治疗组。采用线栓法制备大鼠大脑中动脉闭塞((middle cerebral artery oc-clusion,MCAO)再灌注模型,应用TTC染色检测脑梗死体积,免疫组织化学法检测海马CA1区Bcl-2、Bax蛋白的表达。结果缺血组大鼠海马CA1区Bcl-2、Bax蛋白的表达较假手术组明显增加(P<0.01),给予参附注射液处理后,Bcl-2蛋白表达明显增加(P<0.01),Bax蛋白表达减少(P<0.05),差异均有统计学意义。结论参附注射液可以通过调节Bcl-2、Bax蛋白表达来抑制细胞凋亡,这可能是其发挥脑保护作用的机制之一。

| Abstract: | Objective To study the effect of Shenfu injection on the expression of Bcl-2,Bax in rats with cerebral ischemia-reperfusion.Methods The 30 male Sprague-Dawley rats were divided into 3 groups:sham operation group,cerebral ischemia group and Shenfu injection intervented group.The model rats were established by the middle cerebral artery occlusion(MCAO) with Longa’s methods.The infarct volumes were studied by TTC staining,and immunohistochemitry method were used to detect the expression of Bcl-2,Bax.Results Compared with the sham operation group,the expression of Bcl-2,Bax increased obviously in cerebral ischemia group(P<0.01).After gived Shenfu injection,the expression of Bcl-2 protein increased markedly(P<0.01),and the expression of Bax protein reduced(P<0.05).Conclusion Through regulating the expression of Bcl-2 and Bax protein,Shenfu injection inhibits apoptosis after the transient cerebral ischemia-reperfusion. |
