芹菜甲素的药理作用

本文以合成的dl-芹菜甲素为代表,观察芹菜甲素的系统药理作用。在小于TD₅₀剂量下,芹菜甲素能对抗ⅳ戊四唑所致的全身强直性惊厥,但不能防止EEG癫痫样放电,芹菜甲素对抗脑室注射谷氨酸诱发惊厥、ACh诱发震颤和ip震颤素震颤。芹菜有较强的解除ACh和BaCl₂离体回肠痉挛作用。芹菜甲素对动物呼吸、循环和泌尿系统无明显作用。局部和全身给药也不影响瞳孔和对光反射。

THE PHARMACOLOGICAL ACTIONS OF 3-N-BUTYLPHTHALIOE(AG-1)

The present paper reports the central and peripheral actions of d1-3-n-butyl-phthalide in animals. Beside the anticonvulsant action, Ag-1 was also found to be able to protect mice and rats from seizures induced by sodium glutamate, spasm induecd by i.c.v, acetycholine and tremorine spasm as well as electroshock. No change of EEG patterns was shown in 13 unanesthetized normal rabbits after ip injection of Ag-1 50, 120, 200 and 250 mg/kg. After a single iv of metrazol 20 mg/kg to rabbits, bursts of spike waves were recorded, associated with violent general convulsions. When Ag-1 was injected 30 min prior to metrazol, a similar effect on EEG patterns was recorded, but no behavioral seizure was observed.This compound was also found to have a direct action on acetylcholine or barium chloride induced spasm of isolated rat ileum. Ag-1 in the concentration of 9.5×10^-7M was shown to be able to antagonize the spasmodic action of acetylcholine (2×10^-9 M) and barium chloride (2×10^-6 M) on the rat ileum.
