刺五加苷B、苷E在体外大鼠肠道菌群中的代谢

摘    要:目的:观察体外大鼠肠道菌群对刺五加苷B、苷E的代谢。方法:收集大鼠新鲜粪便在厌氧培养基37℃培养24h,分别加入刺五加苷B、刺五加苷E,培养后加甲醇提取离心,取上清液采用HPLC及UPLC/MS方法对代谢成分进行分离和定性分析。结果:刺五加苷B、苷E在大鼠粪便孵育液中代谢,24h后样本中能检测出较高浓度代谢物。在离体条件下,刺五加苷B、苷E可以被大鼠的肠道菌群代谢,经过UPLC/MS的检测,刺五加苷B代谢物的分子离子峰[M+H]~+为193.08,推测为刺五加苷B的苷元再脱一分子水;刺五加苷E代谢物的分子离子峰[M+H]~+为417.17,推测为刺五加苷B的苷元。结论:刺五加苷B、苷E可以被大鼠肠道菌群代谢。

Abstract:Objective:To study and compare the metabolism of eleutheroside B and E in the rat intestinal flora in vitro.Method: After the rat fresh feces was collected and anaerobically cultured for 24 h at 37℃,eleutheroside B and E was added and cultured,respectively. After extracted by methnol,the main metabolites of eleutheroside B and E were separated and analyzed by HPLC and UPLC/ MS.Result:In vitro,eleutheroside B and E were easily metabolized by the intestinal flora and the main metabolites in incubation medium were in high concentration after 24 h.The metabolite of eleutheroside B with molecular ion peak[M+H]^+ of 193.08 was identified to be its aglycon with a molecular water dehydrated,and the metabolite of eleutheroside E with molecular ion peak[M+H]^+ of 417.17 was identified to be its aglycon.Conclusion:In vitro,eleutheroside B and E were metabolized by the flona.
