加工前、后的亳州牡丹皮水提物对2型糖尿病小鼠血糖、血脂等的作用研究

摘    要:目的:研究加工前、后的亳州牡丹皮水提物对2型糖尿病(T2DM)小鼠的血糖(GLU)、总胆固醇(TC)、三酰甘油(TG)、超氧化物歧化酶(SOD)、丙二醛(MDA)和游离脂肪酸(NEFA)的作用比较。方法:采用高脂饲料喂养雄性昆明小鼠1个月,再采用链脲佐菌素(STZ)120 mg·kg-1进行腹腔注射,复制T2DM小鼠模型,然后,随机分配已建立成功的小鼠模型至模型对照组、阳性对照组、未加工丹皮给药组和已加工丹皮给药组,并以正常小鼠作为正常对照组。对造模成功的小鼠以各丹皮水提物进行28 d的持续灌胃,再解剖取血,测定T2DM小鼠血清中的各项生化指标。结果:已加工牡丹皮和未加工牡丹皮水提物均能显著的降低T2DM小鼠体内的GLU(P<0.01)、TC(P<0.01)、MDA(P<0.05),并能使T2DM小鼠体内的SOD显著升高(P<0.01)。结论:加工前、后的牡丹皮水提物均能显著降低T2DM小鼠的血糖值,两者差异无统计学意义,并对血脂的代谢和抗氧化的能力在一定程度上有调节作用。已加工的牡丹皮外观上虽然更加美观,但是在药效上并未增强。

Abstract:Objective: To study the effects of pre- and post-processed Bozhou cortex moutan aqueous extract on the blood glucose (GLU) , total cholesterol ( TC ), triglycerides ( TG ), superoxide dismutase ( SOD ), malondialdehyde aldehyde ( MDA ) and free fatty acids (NEFA) in type 2 diabetic (T2DM)mice. Method: Kunming male mice were fed with high fat diet for a month followed by streptozotocin (STZ) 120 mg· kg^-1 intraperitoneal injection to obtain type 2 diabetic model, and then randomly divided into the control group,positive control group,the raw cortex moutan-treated group and processed cortex moutan-treated group with normal mice as the normal control. The mice in processed cortex moutan-treated group were orally treated by cortex moutan water extract for 28 days continuously, and then the blood samples were w'ithdrawn to determine the serum biochemical indicators. Result: Processed and unprocessed cortex moutan aqueous extract both could significantly reduce GLU (P 〈0. 01 ) ,TC (P 〈0. 01 ) and MDA (P 〈0. 05) while increase SOD ( P 〈 0. 01 ) in type 2 diabetic mice. Conclusion : Preand post-processed cortex moutan aqueous extract can significantly reduce the blood glucose levels in type 2 diabetic mice without significant difference, and adjust the lipid metabolism and antioxidant capacity to some extent. Cortex moutan processed exhibits more promising appearance, however does not enhance the efficacy.
