摘    要:目的 研究3种目前国内常用的器官保存液对供肝细胞凋亡的影响。方法 分别用UW液、HC-A液和WMO-1号液灌洗并保存大鼠肝脏,于保存后0、12、24h用原位末端标记法检测供肝细胞凋亡情况,并将保存24h的肝及行大鼠原位全肝移植,观察受者的3d存活率。结果 3种保存液保存的肝脏均在保存12h时出现细胞凋亡,HC-A液级瑟WMO-1号液组的凋亡指数(AI)高于UW液组(P〈0.001),而HC-A液组

The effect of different organ storage solutions on apoptosis in rat liver preservation
Abstract:Objective To study the effect of 3 different organ storage solutions commonly used on donor hepatocytic apoptosis. Methods The donor livers were separately flushed and preserved for 24?h with UW, HC A and WMO 1 solutions. The orthotopic liver transplantation in(OLT) rats were performed and the survival rate within 3 days were observed. The apoptosis of donor liver in different time points of 3 storage solutions was detected by in situ terminal labeling technique (TUNEL method). Results The apoptosis occurred 12?h after the liver preservation with 3 storage solutions. The apoptotic index (AI) in HC A and WMO 1 groups was higher than UW group ( P < 0.001 ). In UW preserved group, the AI was increased rapidly following warm oxygenated reperfusion of liver graft preserved in UW solution for 24?h after transplantation. 3 days survival rate of OLT rats in UW group was 60%, significantly higher than in HC A and WMO 1 groups ( P < 0.01 ). Conclusions There might be some difference in the constitu tion between the UW solution and HC A or WMO 1 solutions. The critical factor that caused the apoptosis of transplanted liver cell might be the oxygenated reperfusion following the cold ischemia, and that was the main reason for primary graft nonfunction following liver transplantation. But the injury to hepatocytes caused by apoptosis and difference of organ storage solutions was also one of the important factors that couldn't be neglected.
