摘    要:目的通过研究各类宫颈病变中ETS1的表达,探讨ETS1与宫颈癌发生、发展的相关性。方法①采用总RNA的提取,设计合成引物进行反转录聚合酶链反应(RT-PCR),选用已知的干扰磷酸甘油醛脱氢酶(GAPDH)作为内参照,对不同宫颈组织:健康宫颈组织、宫颈上皮内瘤样病变(CIN)Ⅲ、未发生淋巴结转移的宫颈癌、发生淋巴结转移的宫颈癌,研究ETS1的表达。②进行核蛋白的提取,应用蛋白质印迹法(Western-blot-ting),分析蛋白水平上ETS1在不同宫颈组织中的表达。结果RT-PCR结果显示:ETS1在健康宫颈组织中表达较低,随着向CINⅢ、宫颈癌演变,ETS1的表达量逐渐增加(P<0.05)。在发生淋巴结转移的宫颈癌中表达比未发生淋巴结转移的宫颈癌中的表达高(P<0.01)。Western-blotting结果显示与mRNA水平表达一致。结论ETS1与宫颈癌发生、发展有相关性,而且与肿瘤浸润和转移有密切的关系。

Abstract:Objective To explore the expression of ETS1 in various cervical lesions and its correlation with de- velopment and progression of these conditions. Methods (1)Total RNAs were extracted from tissue samples, and ETS1 genes were amplified by RT-PCR with specially designed primers. The expression of ETS1 gene was study at the mRNA level in four categories of cervical tissue (normal tissue, CIN Ⅲ, cervical cancers with and without lymph node metasta- sis), using GAPDH as the house-keeping gene. (2)Westem-blotting was used to study the expression of ETS1 gene at the protein level. Results RT-PCR assay showed low expression of ETS1 in normal cervical tissue, however, the expression increased with progression from CIN Ⅲ to cervical cancer. Higher expression was detected in lesions with lymph node metastasis than in those without. Western-blotting indicated results compatible to mRNA study. Conclusion ETSI may have an important role in development of cervical cancers, and may be closely related to invasiveness and metastasis of the tumor.
