环氧化酶-2、bcl-2与bax在胃癌的表达及意义

摘    要:目的 研究环氧化酶-2(cox-2)与bcl-2、bax在胃癌中的表达及其与胃癌细胞凋亡的关系。方法 采用SP免疫组织化学法检测胃癌及癌旁组织各60例标本中cox-2、bcl-2、bax的表达,用脱氧核糖核酸末端转移酶介导的dUTP缺口末端标记法(TUNEL)检测胃癌及癌旁组织的凋亡率。结果 胃癌组织的cox-2、bcl-2的表达明显高于癌旁组织(P<0.05),bax的表达明显低于癌旁组织(P<0.05),cox-2阳性组胃癌组织的凋亡率显著低于阴性组;cox-2的表达与bcl-2呈正相关,与bax呈负相关。结论 cox-2、bcl-2、bax的异常表达与胃癌细胞凋亡抑制有关;cox-2能够上调bcl-2的表达并下调bax的表达使胃癌细胞凋亡抑制。

Abstract:Objective To study the expression of cyclooxygenase|2(cox|2), bcl|2 and bax gene in human gastric carcinoma tissues and the relationship between cox|2, bcl|2, bax and the apoptosis of gastric carcinoma cells. Methods S|P immunohistochemistry was used to detect the expression of cox|2, bcl|2 and bax in 60 cases of gastric carcinoma and their adjacent noncancerous tissues, and terminal deoxynucleotidyl transferase|mediated dUTP nick end labeling was used to detect the apoptosis in above tissues. Results The positive rates of cox|2 and bcl|2 in gastric carcinomas were significantly higher than those in the adjacent noncancerous tissues(P<0 05). The positive rate of bax in gastric carcinomas was significantly lower than the adjacent noncancerous tissues(P<0 05). The index of apoptosis in gastric carcinomas was significantly lower than the adjacent noncancerous tissues(P<0 05). Conclusion The apoptosis of gastric carcinoma cells correlated with the expression of cox|2, bcl|2 and bax. The high expression of cox|2 up|regulation the expression of bcl|2 gene and down|regulation of bax expression made the inhibition of apoptosis of gastric carcinoma cells.
