高半胱氨酸SAM-SOD修饰电极的电化学行为

摘    要:目的 研究SOD在高半胱氨酸SAM膜电极上的电化学行为,为进一步实现酶的直接电化学提供依据。将有助于生物传感器的进一步开发与研究。方法 在金基底电极上自组装上高半胱氨酸,在高半胱氨酸SAM膜电极上通过EDC接上SOD,利用循环伏安法研究该修饰电极的电化学行为。结果 高半胱氨酸SAM-SOD修饰电极呈现出了良好的伏安响应,但在含有过氧化氢的溶解中SOD的阴极峰电流呈现明显下降趋势。结论 高半胱氨酸对SOD的电子转移反应具有良好的促进作用。过氧化氢使SOD的电化学响应的可逆性变差。

Abstract:Objective In order to provide data and evidence of the direct electron transfer of enzyme, Electrochemical behaviors of homocysteine SAM-SOD modified electrode in phosphate buffer solution were studied,and this will help the further development of sensors.Methods self assembled monolayer of homocysteine was formed on gold electrode by inserting the gold electrode in phosphate buffer solution of homocysteine,and SOD was added on the SAM modified electrode with EDC.Cyclic voltammetry was used to study the electrochemical behaviors of the modified electrode.Results The results indicated that the quasi-reversible electrochemical behavior was observed on the SAM-SOD modified electrode,and the cathodic peak current of SOD decreased distinctly in H 2O 2 solution.Conclusion Homocysteine is an effective promoter for the electeon transfer of SOD and H 2O 2 can make the cathodic peak current of SOD decrease.
