蒿秦通痹胶囊对胶原诱导性关节炎大鼠治疗作用的研究

目的采用胶原诱导大鼠关节炎模型探讨蒿秦通痹胶囊的治疗作用。方法胶原诱导大鼠关节炎模型造模2周成功后分为模型组、西药对照组、中药对照组及蒿秦通痹胶囊各给药组(大、中、小剂量组)。各组造模第15天开始给药,28 d内定期测定关节肿胀;28 d后处理大鼠,踝关节甲醛液固定,脱钙切片HE染色观察病理变化。结果大鼠关节的肿胀从造模第18天开始消退,至第21天与模型组比较,各组差异均有统计学意义(P0.05,P0.01)。以蒿秦通痹胶囊大剂量组为最好,优于中药对照组和西药对照组;从药物减轻关节肿胀程度上,蒿秦通痹胶囊中剂量组作用稍优于中药对照组,优于西药对照组。病理观察结果提示,蒿秦通痹胶囊各给药组大鼠滑膜水肿、炎性细胞浸润、滑膜增生、血管翳生成、软骨破坏等病理损害均得到改善,其总体改善水平与西药对照组相似,优于中药对照组,且呈剂量相关性。结论蒿秦通痹胶囊能有效治疗胶原诱导大鼠关节炎,对滑膜水肿、炎性细胞浸润、滑膜增生、血管翳生成、软骨破坏等病理损害效果明显。

Study of the Therapeutic Effect on Collagen-Induced Arthritis Treated with Haoqin Tongbi Capsules in Rats

Objective To explore the therapeutic effect of Haoqin Tongbi Capsules on collagen- induced arthritis in the rat models. Methods In 2 weeks of successful modeling of collagen- induced arthritis rat models,a model group,a western medicine control group,a Chinese medicine control group and Haoqin Tongbi Capsules groups（ large- dose,middle- dose and small- dose groups） were divided. The medication started on the 15th day of each model group. The joint swelling was determined regularly within 28 days. The rats were sacrificed after 28 days. The ankle joint was fixed with formalin. The decalcified slices were stained with HE for the observation of pathological changes. Results Joint swelling started relieving on the 18th day after modeling and it was compared with the model group on the 21stday. The difference was significant in each group（ P〈0. 05,P〈0. 01）. The result in the large- dose group was the best,superior to Shirebi group and Prednisone group. Regarding the alleviation of joint swelling,the effect in the middle- dose group was slightly better than that in Shirebi group and better than that in Prednisone group. The pathological observation results suggested that Haoqin Tongbi Capsules groups received the improvements in the pathological damages such as synovial edema,inflammatory cell infiltration,synovial hyperplasia,pannus formation and cartilage destruction. The overall improvements were similar to those in the western medicine control group,better than those in the Chinese medicine control group,indicating dose correlation. Conclusion Haoqin Tongbi Capsules treat effectively synovial edema,inflammatory cell infiltration,synovial hyperplasia,pannus formation and cartilage destruction in the rats of collagen- induced arthritis.
