乳腺浸润性导管癌Survivin及Survivin mRNA表达研究

目的检测Survivin在乳腺浸润性导管癌病变中的表达,探讨其与乳腺浸润性导管癌病变的关系。方法在48例乳腺浸润性导管癌病变组织和其癌旁组织中分别应用原位细胞凋亡检测(TuNEL)试剂盒测定原位细胞凋亡指数(Apoptosis Index,AI),应用免疫组化法检测Survivin,应用原位杂交法检测Survivin mRNA表达。结果Survivin及Survivin mRNA在乳腺浸润性导管癌中表达显著升高(P=0.0016,P=0.0000);Survivin及Survivin mRNA灰度值与AI呈正相关(r=0.533,P=0.005;r=0.455,P=0.003)。结论Survivin及Survivin mRNA表达升高可能参与乳腺浸润性导管癌的发生发展过程,检测Survivin及Survivin mRNA对于乳腺浸润性导管癌与良性组织判别具有临床参考意义。

Significance of Survivin and Survivin mRNA in breast carcinoma

Objective To study the relationship between Survivin and breast carcinoma by combining assays of Survivin and Survivin mRNA in breast tissue.Methods Apoptosis Index(AI)was evaluated by TUMEL method,the expression of Survivin and Survivin mRNA were detected by immunohistochemistry and In Situ Hybridization in 48 breast carcinoma cases and those tissues beside carcinoma.Results The higher level of Survivin and Survivin mRNA were found in breast carcinoma compared with tissues beside carcinoma(P=0.001 6,P=0.000 0).There was positive relationship between the expression of Survivin,Survivin mRNA and AI(r=0.533,P=0.005;r=0.455,P=0.003).Conclusion Survivin may participate in proceeding of breast carcinoma,and it is suggested that combined detection of Survivin and Survivin mRNA may have important referent significance on the differentiation of benign and malignant breast tissue.
