摘    要:目的研究ATP对肾脏髓袢升支粗段管周膜钾通道活性的调控作用。方法应用单通道膜片钳技术记录管周膜钾通道电流,观察ATP作用前后钾通道开放概率的变化。结果在内面向外式记录模式下,1.0mmol·L-1的ATP对管周膜50pS钾通道有抑制作用,通道开放概率从加药前的0.46±0.28降至0.02±0.04;ATP对50pS钾通道的抑制作用具有剂量依赖性。结论髓袢升支粗段管周膜钾通道的活性受细胞内ATP的调控。

Abstract:Aim To investigate the effect of ATP on the basolateral K + channels in the thick ascending limb(TAL) of rat kidney.Methods The single channel patch-clamp technique was used to record the basolateral K + channel current in the TAL,and channel open probability was calculated before and after the addition of ATP.Results 1mmol·L -1 ATP inhibited 50 pS K + channels in basolateral membrane of TAL in insideout patches,and channel open probability was decreased from 0.46±0.28 to 0.02±0.04 after addition of ATP.The inhibitory effect of ATP on 50 pS K + channels was dose-dependent.Conclusion The activity of K + channel in basolateral membrane of TAL is regulated by cellular ATP.
