食管癌细胞原代培养体外药敏试验研究

目的探讨(MTT)法体外药敏试验对贲门癌食管癌术后化疗的指导价值。方法采用MTT法检测60例食管癌标本对临床常用的五种化疗药物单独及联合应用的体外敏感性。结果单药组组问有显著性差异(x2=127.324,P<0.001),x2检验证实五种抗癌药物单用时最敏感的药物为CPT,敏感率为85％;联合用药组的总体敏感率明显高于单一用药组的总体敏感率,x2=63.569,P<0.001)。结论MTT法体外药敏试验指导食管癌术后的化疗可灵活的对患者进行针对性较强的个体化治疗,以减少用药的盲目性和因盲目给药所造成的不良反应,从而提高疗效,增加生存率并改善生存质量。

Clinical evaluation of chemosensitivity testing for postoperative patients with esophageal cancer using MTT assay in vitro

Objective To study the predictive value of MTT in vitro assay for chemotherapy to esophageal cancer patients.Methods Primary culture of tumor cells taken from 31 cases of esophageal cancer were performed with chemosensitivity testing by MTT assay in vitro.Results Among the 5 commonly used antineoplastic agent include in this study,CPT is the most sensitive drugs when used along in chemosensitivity testing in vitro.While 5-Fu plus CPT and CPT plus MMC shows more effectivity than othes in the combination group.The combined group shows more sensitivity than that of the group which used drugs alone(x2=63.569,P<0.001).Conclusion MTT in vitro assay appears to be a good way for predicting the drug sensitivity and resistance which identifies chemosensitive patients and which may allow some individualization of chemotherapy.
