芪仙汤对支气管哮喘小鼠肺组织FIZZ1 mRNA及蛋白表达的影响

摘    要:目的:观察芪仙汤对哮喘小鼠肺组织中FIZZ1基因及蛋白表达水平的影响,探讨芪仙汤治疗哮喘的作用机制。方法:选用30只健康雌性BALB/c小鼠,采用随机数字表分为正常对照组(对照组)、支气管哮喘模型组(模型组)、芪仙汤治疗组(治疗组)。除对照组外均采用卵白蛋白(OVA)建立支气管哮喘小鼠模型。各组分别相应干预灌胃2周。从小鼠的一般情况、肺组织病理形态学(HE染色病理切片)指标评价支气管哮喘小鼠模型;采用RT-PCR检测肺组织FIZZ1基因mRNA表达水平,Western blot和免疫组化法检测肺组织FIZZ1蛋白表达水平。结果:HE染色结果显示,模型组小鼠肺组织较正常组嗜酸性粒细胞等炎症细胞浸润明显,气道壁及支气管平滑肌增厚明显;治疗组哮喘病理改变明显改善。RT-PCR、Western blotting、免疫组化结果均显示模型组FIZZ1表达水平显著高于对照组,治疗组小鼠肺组织中FIZZ1表达水平明显低于模型组,差异均有统计学意义(P0.05,P0.01)。结论:芪仙汤可能是通过下调小鼠肺组织FIZZ1表达起到治疗支气管哮喘的作用。

Abstract:  Objective:To observe the effects of Qixian Decoction on gene and protein expression of found in inflammatory zone 1(FIZZ1) in lung of mice with asthma,and explore the mechanism of Qixian Decoction in the treatment of asthma.   Methods: Thirty healthy female BALB/c mice were selected and randomly divided into the normal control group(control group),model group of bronchial asthma(model group)and Qixian Decoction group(treatment group)according to random number table. Except the control group,the mouse model of bronchial asthma was established by ovalbumin (OVA) in other mice. The corresponding intervention was given in each group by intragastric administration for 2 weeks. The mouse model of bronchial asthma was evaluated by the general condition and the pathological morphology of lung tissue after HE staining. The mRNA expression of FIZZ1 in lung tissue was detected by RT-PCR,and the protein expression of FIZZ1 in lung tissue was detected by Western blot and immunohistochemistry.   Results: Compared with the normal control group,the obvious inflammatory cells infiltration such as eosinophils was found in lung tissue of mice in the model group after HE staining,and there were obvious thickening of airway wall and bronchial smooth muscle. The pathological changes of asthma in the treatment group were obviously improved. The results of RT-PCR,Western blot and immunohistochemistry all showed that the expression level of FIZZ1 in the model group was significantly higher than that in the control group,and the expression level of FIZZ1 in lung tissue of mice in the treatment group was significantly lower than that in the model group,with statistical differences(P<0.05,P<0.01).   Conclusion: Qixian Decoction may play a role in treating bronchial asthma by down-regulating the FIZZ1 expression in lung tissue of mice.
