柴郁地仙方对围绝经期抑郁症模型大鼠行为学及HPA轴的影响

摘    要:目的:探讨中药复方柴郁地仙方治疗围绝经期抑郁症(PDD)的作用机制。方法:48只3~4月龄雌性SD大鼠随机分为假手术组、模型组、西药组及柴郁地仙方高、中、低剂量组,每组8只。假手术组不予去势,其余组行卵巢去势术后孤养并予以28 d慢性不可预见性应激建立PDD模型,造模同时各组分别予柴郁地仙方、盐酸氟西汀和戊酸雌二醇混悬液及0.9%Na Cl溶液连续灌胃28 d。观察应激过程中大鼠行为学及体质量变化;采用酶联免疫吸附测定法(ELISA)检测血清中促肾上腺皮质激素释放激素(CRH)、促肾上腺皮质激素(ACTH)、皮质酮(CORT)含量。结果:模型组大鼠体质量增长缓慢,旷场水平得分及糖水偏好率相对于假手术组均明显降低(P0.01);血清中CRH、ACTH、CORT含量较假手术组明显升高(P0.01)。与模型组比较,柴郁地仙方高、中、低剂量组大鼠旷场水平得分、糖水偏好率及体质量均明显升高(P0.01),血清中的CRH、ACTH、CORT含量明显降低(P0.05,P0.01)。结论:柴郁地仙方可能通过下丘脑-垂体-肾上腺轴的调节作用而起到治疗围绝经期抑郁症的作用。

Abstract:  Objective:To discuss the mechanism of Chaiyu Dixian Decoction in the treatment of perimenopausal depressive disorder (PDD).   Methods: 48 female SD rats that 3-4 months old were randomly divided into the sham group,model group,western medicine group and Chaiyu Dixian groups with high-,medium- and low-dose,8 rats in each group. The sham group received no ovariectomy,the other groups underwent ovariectomy and separate feeding,and were treated with chronic unpredictable mild stress for 28 days to establish PDD model, meanwhile, each group was treated with Chaiyu Dixian Decoction, fluoxetine hydrochloride, estradiol valerate suspension and 0.9%NaCl solution by intragastric administration for continuous 28 days, respectively. The changes of behavior and body mass in rats during stress process were observed. The serum contents of corticotropin releasing hormone(CRH),adrenocoticotropic hormone(ACTH)and corticosterone(CORT)were detected by enzyme linked immunosorbent assay(ELISA).   Results: Compared with the sham operation group,the body mass of rats in the model group was increased slowly,the scores of open field test and sucrose preference were obviously decreased(P<0.01),the serum contents of CRH,ACTH and CORT were obviously increased(P<0.01). Compared with the model group,the body mass and the scores of open field test and sucrose preference were obviously increased in the Chaiyu Dixian groups with high-,medium- and low-dose(P<0.01),and the serum contents of CRH,ACTH and CORT were obviously decreased(P<0.05,P<0.01).   Conclusion: Chaiyu Dixian Decoction may play the role in the treatment of perimenopausal depressive disorders through the regulation of hypothalamus-pituitary-adrenal axis.
