内皮素在梗阻性黄疸大鼠血液、肝、肾、胃及肺组织中的含量变化和作用研究

摘    要:为了解大鼠胆管梗阻后内皮素在肝、肾、胃、肺组织及血液中的分泌状况以探讨内皮素在梗阻性黄疸中的作用。用放射免疫法(Radioimmunoassay.RIA)测量大鼠胆管梗阻后5、10、15、20、25d时肝、肾、胃、肺组织及血液中内皮素的含量以及肝组织、血液中甘胆酸的含量,组织切片观察上述组织的病理改变。血液及肝组织中内皮素和甘胆酸含量呈进行性升高,且二者升高水平呈正相关(P<0.05)。肾、胃、肺组织中内皮素含量亦相继升高(P<0.05)。病理组织切片显示随胆管梗阻时间延长上述组织均呈进行性损害,提示内皮素可能是参与梗阻性黄疸肝脏损伤的重要因素之一,并且对梗阻性黄疸后多系统器官衰竭的触发可能具有重要的病理生理意义。

Abstract:In order to observe the secretion of endothelin in liver,kidney,stomach,lung tissues andblood after ligation of the common bile duct and to evaluate the role of endothelin in obstructive jaundice, the concentration of endothelin in tissues of liver, kidney, stomach, lung andblood and that of cholyglycin in liver and blood were determined 5, 10, 15, 20 and 25 days respectively after ligation of common bile duct by radioimmunoassay. The histopathologicalchanges of these tissues were examined. The results showed that concentrations of endothelinand cholyglycin in blood and liver elevated progressively and correlated positively (P<0.05). The concentrations of endothelin in kidney,stomach and lung tissues also elevated (P<0.05). Histological examination showed that the above tissues were damaged progessivelywith prolonged ligation of the bile duct. It is suggested that endothelin may be an importantfactor leading to damage of liver in obstructive jaundice,and may have important pathophysiological significance in triggering multisystemic failure after obstructive jaundice.
