脑缺血后大鼠大脑新皮质的nestin表达与神经节苷脂GM1的关系

摘    要:目的观察大鼠缺血脑损伤后,外源性单唾液酸四己糖神经节苷脂(GM1)与大脑新皮质巢蛋白(nestin)的表达的关系。方法Wistar成年大鼠随机分配到GM1组、对照组和假手术组;建立大脑中动脉永久梗死性模型,腹腔注射GM1后,利用免疫组织化学染色方法,观察模型大鼠大脑新皮质的巢蛋白表达。结果(1)GM1组表达nestin的细胞数明显比对照组和假手术组多,对照组又明显比假手术组多;(2)对照组表达nestin的细胞数是在干预后7d达最高,干预后10d与3d相比,无差别;GM1组则是干预后7d达高峰,10d后下降,但仍比3d的高。结论GM1能促进缺血脑组织的nestin表达的增加。

Abstract:Objective To research the relationship between expression of nestin in the adult rat neocortex and monosialoteterahexosyl ganglioside (GM1) after cerebral schemia.Methods Wistar rats wcre randomly as- signed to three groups.GM1,control and sham operated group.By using rat permanent middle cerebral artery oc- clusion as a model and intraperitoneal injection GM1,immunohistocbemical staining was used to observe ncstin pro- tein of neocortex.Results (1)Nestin positive cells in the GM1 group were significantly higher than the control group and the sham operated group;(2)The nestin positive cells of the control group and the GM1 group all reached maximal expression on the seventh day,but no significant difference between the third day and the tenth day in the controls,while the cells on the tenth day's were significantly higher than the third day's in the GM1 group.Conclusion The GM1 can promote the expression of nestin in cercbral ischemia.
