电针内关对心肌缺血再灌注损伤大鼠腺苷及bcl-2基因表达的影响

摘    要:目的:通过研究电针内关对心肌缺血再灌注损伤过程中内源性保护物质腺苷(Ado)及细胞凋亡调控基因bcl-2的影响,阐明电针内关抑制心肌缺血再灌注损伤的作用机理。方法:在大鼠冠状动脉左前降支根部穿线建立心肌缺血再灌注损伤模型,电针内关穴,检测血清中腺苷含量,分析凋亡细胞基因的表达。结果:内关穴组血清Ado含量高于模型组(P<0.01);模型组bcl-2蛋白表达的阳性率较低,电针内关组阳性表达率明显增高,与模型组比较均有非常显著性意义(P <0.01)。结论:电针内关穴可促进内源性保护物质腺苷的释放,且可对凋亡的调控基因bcl-2的生成有促进作用,从而在一定程度上对心肌缺血再灌注损伤心肌起到保护作用。

Abstract:Objective:To explore the effects of electroacupuncture in Neiguan points on adenosine and the genes expression of bcl-2 during the process of ischemia and reperfusion injury so as to explain the protective mechanism of acupuncture. Methods: The rats were established as the myocardial ischemia and reperfusion injury models by ligating the left anterior descending branch root of the coronary artery, Neiguan points were be electroacupunctured. Then, the contents of adenosine were detected and the expression of bcl-2 was observed. Results: The contents of adenosine in the Neiguan group was higher than that in model group(P<0.01). The positive rate of bcl-2 expression was lower in the model group and higher in acupuncture groups which was very significant compared with model groups (P<0.01). Conclusion: Electroacupuncture of Neiguan points can accelerate the release of adenosine and influence the genes expression of bcl-2, reduce the injury degree during the process of ischemia and reperfusion injury and protect cardiac cell.
