摘    要:骨质疏松症是中老年人的常见病、多发病,严重地危害老年人的健康。相比于骨密度检测而言,骨转换标志物监测可更早地评估骨质疏松疗效,还可早期发现对于治疗无应答的患者。骨钙素是一种灵敏度较高的骨转换标志物,其含量可以反映成骨细胞活性,同时也反映了骨吸收和骨重构的情况。因此文章构建了一种用生物素-链霉亲和素系统连接的双纳米金侧流免疫层析试纸条定量检测骨钙素的方法。骨钙素的检测限是0.1ng/mL,线性范围为0.1~200ng/mL,批内和批间的平均回收率分别为96.4%~108.4%和99.3%~107.5%,相应的变异系数分别为5.3%~8.6%和4.9%~7.7%。

Abstract:Osteoporosis is a common disease among elderly people,which endangers the health of the elderly seriously.Compared with bone mineral density,bone turnover markers(BTMs)can quickly reflect the therapeutic effect of osteoporosis and discover patients with no response to treatment.Osteocalcin(OCN)is a kind of BTMs that had high sensitivity,and by measuring the amount of OCN in blood,the osteoblast activity can be measured and bone resorption and bone remodeling also can be reflected.In this article,the author constructed a lateral flow immunochromatographic assay(LFIA)with dual gold nanoparticles connected by biotinstreptavidin system for quantitative detection of OCN.The limit of detection(LOD)for OCN was 0.1ng/mL with a good linear range of 0.1 ng/mL to 200 ng/mL.The average recoveries for intra-and inter-assay ranged from 96.4%to 108.4%and from 99.3%to 107.5%with corresponding CVs from 5.3%to 8.6%and from 4.9%to 7.7%,respectively.
