MMP2、MMP9和PCNA在大肠肿瘤中的表达及临床意义

目的 研究MMP2、MMP9及PCNA在大肠肿瘤中的表达及临床意义。方法 运用免疫组织化学S-P法检测52例大肠癌组织、15例大肠腺瘤及10例正常大肠粘膜组织的MMP2、MMP9和PCNA表达。结果(1)MMP2、MMP9和PCNA的阳性表达强度及阳性表达率(PCNA为强阳性表达率)按正常组织、大肠腺瘤、大肠癌的顺序依次增高，且表达强度和阳性率(PCNA为强阳性率)在三组间均有显著性差异；(2)大肠癌组织中MMP2表达和MMP9表达间、MMP2表达和PCNA表达闻、MMP9表达和PCNA表达间均有明显相关性，(3)PCNA表达与大肠癌的分化程度、有无淋巴结转移密切相关，分化程度越低，PCNA表达越强，发生淋巴结转移的机率越大。结论 MMP2、MMP9的过度表达有助于大肠癌的恶性发展，PCNA表达与大肠癌的分化程度、淋巴结转移及临床分期密切相关，是评估大肠癌细胞增殖活性和恶性程度的可靠指标，大肠癌细胞的增殖活性与其MMP2、MMR9的分泌有一定关系。

Expression of MMp2、MMP9 and PCNA in colorectal tumors and its clinical implication

Objective To investigate the expression of MMP 2? MMP 9 and PCNA in colorectal tumors, and to discuss their effect on the progress of colorectal carcinoma. Methods The immunohistochemical staining with S P method was used to detect the expression of MMP 2?MMP 9 and PCNA in paraffin sections from 52 patients with colorectal carcinoma?15 patients with colorectal adenoma and 10 normal colorectal mucosal tissues .Results (1) The expression intensity and rate of MMP 2? MMP 9 and PCNA(strong expression rate) increased in turn by normal mucosal tissues? colorectal adenomas and colorectal carcinoma, and there were marked differences among them. (2) The expression was markedy correlated between MMP 2 and MMP 9, MMP 2 and PCNA, MMP 9 and PCNA in colorectal carcinoma. (3) The expression of PCNA in colorectal carcinoma was closely associated with differentiation and lymph node metastasis. The poorer the differentiation was, the stronger the PCNA expression was, and it was easier to make metastasis.Conclusion Overexpression of MMP 2?MMP 9 faciliates the malignant progress of colorectal carcinoma; The expression of PCNA is related to differentiation?lymph node metastasis and clinical stage of colorectal carcinoma, it is a reliable marker for proliferative activity and malignant level of colorectal carcinoma; The proliferative activity of colorectal cancer cells correlate with their secretion of MMP 2?MMP 9.
