电针对脑缺血再灌注大鼠海马 Fos蛋白的影响

摘    要:目的:探讨脑缺血再灌注后电针对大鼠海马Fos蛋白表达的影响。方法:SD大鼠18只,体质量250~280g,雌雄不限。动物随机分为3组,对照组、模型组和治疗组。采用夹闭大鼠双侧颈总动脉造成的脑缺血再灌注损伤模型,电针百会、风池、大钟及足三里穴,频率2~20Hz,强度2.0A,持续30min,4h后观察海马Fos蛋白的表达。结果:电针能明显加强脑缺血再灌注后海马各区Fos蛋白的表达。治疗组与模型组相比海马各区阳性细胞数显著增加,CA1区(236±26,126±19,P<0.05);CA3(328±80,212±55,P<0.05);CA4(594±104,381±92,P<0.01);DG(621±111,341±89,P<0.01)。结论:缺血再灌注可诱导海马Fos蛋白的显著表达,电针可增强Fos蛋白的表达。

Abstract:AIM:To access the effects of electro-acupuncture on the expression of Fos protein in rat hippocampus after cerebral ischemia-reperfusion injury. METHODS:Eighteen SD rats were randomly divided into control,model and treatment groups.Bilateral carotid artery occlusion was performed in establishing reperfusion model of focal cerebral ischemia in the latter 2 groups,and electro-acupuncture was administered in the treatment group at the acupoints of " Paihui" ," Fengch" ih," Tachung" and " Tsusanli" at a frequency of 2-20 Hz and the intensity of 2.0 A for 30 minutes.After 4 hours,the expression of Fos protein in rat hippocampus was observed. RESULTS:Electro-acupuncture obviously elevated Fos expression in the hippocampal regions after reperfusion following cerebral ischemia.In the treatment group,Fos-positive cells of all hippocampal regions were significantly increased in comparison with the model group(CA1:236± 26 vs 126± 19,P< 0.05;CA3:328± 80 vs 212± 55,P< 0.05;CA4:594± 104 vs 381± 92,P< 0.01; DG:621± 111 vs 341± 89,P< 0.01). CONCLUSION: Reperfusion after cerebral ischemia in the hippocampus can obviously induce Fos expression that can be strengthened by electro-acupuncture.
