回心康片抗心肌细胞缺氧损伤的作用及机制

摘    要:目的:探讨回心康片抗大鼠心肌细胞缺氧损伤的作用及其机制。方法:建立体外大鼠乳鼠心肌细胞缺氧损伤模型。采用血清药理学方法,研究回心康片对细胞活力的影响;采用全自动生化分析仪检测谷胱甘肽(GSH)、一氧化氮(NO)含量;采用FLIPR Calcium4试剂盒检测药物对胞浆内钙离子浓度变化的影响。结果:回心康片对正常心肌细胞无明显影响,但可明显提高缺氧损伤心肌细胞的活力,降低NO水平,提高GSH活性,降低心肌细胞因缺氧损伤而造成的钙离子浓度升高,与缺氧损伤组比较差异具有统计学意义(P<0.05,P<0.01)。结论:回心康片能抗心肌细胞缺氧损伤,其机制可能与抗氧化和减轻钙超载有关。

Abstract:OBJECTIVE To study the effect of Huixinkan tablets on cardiocytes injured by hypoxia and its mechanism of actions.METHODS Neonatal cardiomycytes were cultured and cytotoxicity in cultured cardiomyocytes was induced by hypoxia.Then,a method of serum pharmacology was used to study the effect of Huixinkan tablets on cell viability,which was analyzed using the MTT assay.GSH and NO activity in medium were assayed by automatic biochemistry analyzer and i of cardiomyocytes loaded by Fluo-4 was analyzed by Multiskan Ascent immediately.RESULTS The tablets showed no significant effect on healthy cardiomyocytes.Huixinkan tablets increased the activities of the cells injured by hypoxia significantly.GSH and NO activities in medium and the intracellular calcium concentration dealed by Huixinkan tablets medicated serum were significantIy different from the model group(P<0.05,P<0.01).CONCLUSION Huixinkan tablets have protective effect on the myocardial cells injuried by hypoxia,its mechanism may be associated with antioxidant and relieving calcium overload.
