摘    要:目的 建立高效液相色谱法测定壮药铁包金中大黄素、大黄酚的含量.方法 采用Diamonsil C18柱(250 mm×4.6 mm,5μm)色谱柱,流动相为甲醇-0.4%(体积分数)H3PO4(体积比75∶25),流速为1.0mL/min,柱温为28℃,检测波长为254 nm.结果 铁包金中大黄素和大黄酚分别在1.4~ 14 μg/mL(r=0.999 6)和6~60 μg/mL(r=0.999 0)范围内线性关系良好,平均加样回收率分别为99.2%、95.8%,RSD值分别为2.0%、2.3%.结论 首次对铁包金药材中蒽醌类成分进行了含量测定,所建立的方法分离度高、方法学验证符合要求,可用于同时测定壮药铁包金中大黄素和大黄酚的质量分数.

Determination of emodin and chrysophanol in Zhuang medicine Berchemia lineata by HPLC

Abstract:Objective To establish an HPLC method for the simultaneous determinations of emodin and chrysophanol in Berchemia lineata. Methods Diamonsil C 18 column (250 minx4.6 mm,5 μm) was used as stationary phase.Mobile phase was methanol-0.4% phosphoric acid (75:25) with the flow rate of 1.0 mL/ rain.The column temperature was 28 ℃ .The detection wavelength was 254 rim.Results The linearity was good in the range of 1.4-14 μg/mL of emodin(r= 0.999 6) and in the range of 6-60 μg/mL(r=0.999 0) of chrysophanol.The average recovery of emodin and chrysophanol was 99.2% with RSD 2.0% and 95. 8% with RSD 2. 3% ( n = 6) , respectively. Conclusion The contents of emodin and chrysophanol in Zhuang medicine B. lineata were measured for the first time. The established method can be used for the determination of emodin and ehrysophanol in B.lineata simultaneously.
