STAT3反义寡核苷酸对HepG2肝癌细胞增殖抑制和凋亡诱导作用

摘    要:目的 探讨STAT3反义寡核苷酸对HepG2细胞增殖的抑制作用和细胞凋亡的诱导作用。方法: HepG2细胞体外培养,人工合成并硫代修饰STAT3 ASODN,通过脂质体转染进入HepG2细胞,倒置显微镜下观察细胞形态学变化、 MTT法检测细胞增殖抑制程度,原位末端标记(Tunel)法检测细胞凋亡指数(AI), RT-PCR检测细胞中STAT3 mRNA 的表达水平,流式细胞仪检测细胞周期的变化和细胞凋亡率。结果: STAT3 ASODN各浓度组对HepG2细胞的增殖有明显的抑制作用,能明显诱导细胞凋亡。结论: STAT3 ASODN能够诱导HepG2细胞凋亡。

Effects of STAT3 antisense oligodeoxy-nucleotid on apoptosis and proliferation of HepG2 cells
Abstract:Objective To investigate the effection cell prolifection and cell apoptosis of STAT3 antisense oligodeoxy-nucleotid on HepG2cells. Methods HepG2 cells were serial subcultured, STAT3 antisense oligodeoxy-nucleotid were transfected into HepG2 cells by lipofectamine mediated transient transfection method. Cell morphological transformation was observed with inverted microscope and cell proliferation was measured by MTT method , Cell apoptosis index was examined by Tunel,RT-PCR was used to make a STAT3 gene expression,FCM was used to make a apoptosis rate and cell cycle. Results STAT3 ASODN had obvious inhibitory effects on the cell proliferation and induced cell apoptosis of HepG2 cells. The cell apoptosis rate in STAT3 antisense oligodeoxy-nucleotid group was obviously higher than control groups。 Conclusion STAT3 ASODN can induce cell apoptosis in HepG2 cells.
