温经通络法对胶原性关节炎大鼠血管新生干预作用的研究

摘    要:目的观察血管内皮细胞生长因子(VEGF)及血管内皮细胞生长因子受体(KDR)在胶原性关节炎(CIA)大鼠血清及滑膜组织中的表达变化,及芍药甘草附子汤的干预作用。方法建立CIA大鼠模型,分组干预后计算关节炎炎症指数(AI)。ELISA法检测血清VEGF、sKDR含量,免疫组织化学法检测关节滑膜VEGF、KDR表达。结果造模后第28天各组大鼠炎症反应明显,用药4周后炎症反应明显好转,对照组和治疗组明显好于模型组(P〈0.05),但两组之间差异无统计学意义(P〉0.05)。模型组病理评分较正常组显著增高(P〈0.01),各用药组光镜下病理改变较模型组减轻。血清VEGF、sKDR含量,模型组明显高于正常组(P〈0.05),用药后对照组和治疗组明显下降(P〈0.05)。免疫组化结果显示,VEGF、KDR均定位于细胞浆,对照组和治疗组的阳性细胞数较模型组明显减少(P〈0.05)。结论芍药甘草附子汤对CIA大鼠有明显的治疗作用,其机制可能与抑制VEGF及其受体表达有关。

Intervention of Angiogenesis in the Rats of Collagen Induced Arthritis Treated with the Therapy for Warming Meridians and Circulating Collaterals

Abstract:Objective To observe the changes in the expressions of vascular endothelial growth factor(VEGF) and vascular endothelial growth factor receptor(sKDR) in the serum and synovial tissue of the rats with collagen induced arthritis ( CIA), as well as the intervention effect of shaoyao ganeao fuzi decoction. Methods CIA rat model was set up. The arthritis inflammatory index (AI)was calculated after intervention in groups. ELISA was adopted to determine the contents of serum VEGF and KDR. IHC assay was applied to determine VEGF and sKDR expressions in joint synovium. Results After modeling,on the 28th clay,the inflammatory reaction was apparent in the rats of each group. In 4 weeks of medication, the inflammatory reaction was relieved apparently. The improvement in the control group and treatment group was superior apparently to the model group( P 〈 0.05 ). But the difference was not significant between the control group and the treatment group(P 〉0.05). The pathological score in the model group was higher significantly than that in the normal group(P 〈0. 01 ). The pathological change was alleviated mieroseopieally in each medication group as compared with the model group. The contents of serum VEGF and KDF in the model group were higher apparently than those in the normal group( P 〈 0.05 ). The contents were reduced apparently after medication in the control group and treatment group( P 〈 0. 05 ). IHC results showed that VEGF and KDR were localized in cellular plas- ma. The number of positive cell in the control group and the treatment group was less apparently than that in the model group (P 〈 0.05 ). Conclusion Shaoyao ganeao fuzi decoction has the apparent therapeutic effect on CIA in the rats. The therapeutic mechanism is possibly relevant with the inhibition of VEGF and its receptor.
