摘    要:摘要] 目的 探讨马拉色菌在直接免疫应答中诱导角质形成细胞表达TLR-2的作用及对sICAM-1和IL-8分泌和表达的影响。方法 马拉色菌和角质形成细胞体外共培养,通过免疫细胞化学方法检测角质形成细胞TLR-2蛋白的表达状况,并运用ELISA方法检测马拉色菌对角质形成细胞sICAM-1和IL-8表达和分泌的调节作用。结果 体外实验发现,在二者共同培养时,马拉色菌能增强角质形成细胞TLR-2的表达,且角质形成细胞的sICAM-1的和IL-8表达和分泌也增加。结论 马拉色菌可诱导TLR-2的增强表达,并能促进sICAM-1和IL-8的表达和分泌,这一机制可能在痤疮等相关的炎症反应中起重要作用。

The effect of Malassezia furfur on expression and production of TLR-2,sICAM-1 and IL-8 by dermal keratinocytes
Abstract:Objectives To investigate the role of Malassezia furfur in immediate immunity by inducing expression of TLR-2 by keratinocytes, and also to study the secretion and expression of sICAM-1 and IL-8 in keratinocytes is induced by Malassezia furfur.Methods Keratinocytes monolayers and Malassezia furfur were co-cultured in vitro, and the modulating effects of Malassezia furfur on the expression of TLR-2 was measured by immuocytochemistry methods, the expression and secretion of sICAM-1 and IL-8 in keratinocytes was detected by enzyme linked immunosorbent assay. Results Compared with the controls, the expression of TLR-2 induced by Malassezia furfur was increased in the keratinocytes, and the expression and secretion of the sICAM-1 and IL-8 also was increased. Conclusions This work demonstrated that Malassezia furfur could induce TLR-2 expression and also promoted the expression and secretion of sICAM-1 and IL-8 in keratinocytes,this mechanism could play an essential role in acne-linked inflammation.
