摘    要:目的 探讨SLE患者外周血单个核细胞 (PBMC)凋亡率异常是否与其自身抗体产生高低有关。方法 采用碘化丙锭 (PI)染色法 ,在流式细胞仪下定量检测 2 0例SLE患者及 10名正常人PBMC体外凋亡情况 ,同时观察了SLE患者自身抗体的表达及与凋亡率的关系。结果 SLE患者PBMC体外培养 0、2 4、48、72h后凋亡率较正常人明显增高 (P <0 .0 5 ) ,且活动期SLE高于非活动期 (P <0 .0 5 )。SLE患者自身抗体产生越多者较自身抗体产生少者的凋亡率明显增高 (P <0 .0 1)。结论 SLE患者PBMC的凋亡加速 ,与其自身抗体产生增高有关 ,可能在SLE的发病机制中起一定作用

Abstract:Objective To investigate whether there is correlation between abnormal rate of peripheral blood mononuclear cell (PBMC) apoptosis and autoantibody level.Methods Using flow cytometry and propidium iodide(PI) staining,the rates of apoptosis in different PBMC from patients with systemic lupus erythematosus (SLE) and from the healthy controls were investigated.The relationship of between the expression of autoantibody and apoptotic rate of PBMC from SLE patients was determined.Results After PBMCs were incubated for 0,24,48,72 hours,the rate of PBMC apoptosis in 20 SLE patients increased significantly compared with that in the 10 healthy controls (P<0.05).And the rate of PBMC apoptosis from active SLE patients were higher than that from inactive SLE patients.The results also showed that the more the production of autoantibodies,the higher the rate of apoptosis (P<0.01).Conclusion The apoptosis of PBMC in SLE accelerates in vitro,by which autoantibodies are produced.The abnormalities of PBMC apoptosis may contribute to the pathogenesis of SLE.
