上皮钙粘蛋白、α-和β-连环蛋白在肝癌组织中的表达

目的　观察细胞粘附与信号转导分子E -Cadherin ,α-actenin和 β-catenin在正常肝组织、肝癌旁肝硬化组织和肝癌组织中的表达和胞内分布变化 ,以探讨肝癌发生、发展的机制。方法　应用免疫组织化学法 (链霉抗生物素 -过氧化物酶显色 )对正常肝组织、癌旁肝硬化组织和肝癌组织进行染色。结果　 2例正常肝组织中表现为上述分子在胞膜表达 ,胞浆不表达。 1 7例癌旁肝硬化组织中上述分子出现胞膜表达增强 ,胞浆也出现表达。 1 7例肝癌组织中上述分子表现为胞膜表达减弱或消失 ,胞浆表达增强 ,且表达的细胞稀少和分布不均。结论　肝癌细胞内粘附和信号转导分子E-Cadherin ,α -catenin和 β-catenin发生转位或再分布现象 ,提示癌细胞在一定程度上丧失粘附和信号转导功能

Expression of E-cadherin, α- and β-catenin in human hepatocellular carcinoma tissues

Objective In order to investigate the mechanism of tumorigenesis and development of human hepatocellular carcinoma, we examined the cell adhesion and signaling molecules: E_Cadherin, α- and β-catenin expression and their cellular localization in human hepatocellular carcinoma. Methods An immunnohistochemistry method was used to immunostain the specimens from normal liver tissues, hepatocirrhosis tissues adjacent to liver cancer and carcinoma tissues. Results E-Cadherin, α- and β-catenin expressed in 2 normal liver tissues at cell membrane but not in cytosol, while in 17 hepatocirrhosis tissues adjacent to liver cancer, membrane expression increased and cytosolic expression had been observed. In 17 hepatocellular carcinoma tissues, the molecules were expressed mainly in cytosol, membrane expression reduced, cells that expressed the molecules were seldom and uneven. Conclusions The cell adhesion and signaling molecules: E-Cadherin, α- and β-catenin in human hepatocellular carcinoma cells had translocalized or relocalized, it suggests that may be the cancer cells lost normal adhesion and signaling function.
