磷酸苯丙哌林在犬体内的药代动力学研究

摘    要:目的建立高效液相色谱法并测定犬血浆中苯丙哌林的浓度,研究磷酸苯丙哌林在beagle犬体内的药代动力学过程。方法 beagle犬单剂量口服80 mg磷酸苯丙哌林,采用高效液相色谱法测定血浆中苯丙哌林的浓度。结果苯丙哌林在beagle犬体内过程符合一室模型,血药峰浓度(Cmax)为(133.52±58.23)ng/mL,达峰时间(Tmax)为(2.96±0.58)h,半衰期(t1/2)为(2.27±0.51)h,0~24 h药时曲线下面积(AUC0~24)为(985.16±135.23)ng.h/mL。结论所建立的高效液相色谱法简便、快速,适用于体内苯丙哌林的浓度测定及药代动力学研究。

Abstract:Objective To establish an HPLC method to detect the concentration of benproperine in beagle dog plasma for studying its pharmacokinetics in vivo. Methods Beagle dogs were orally given benproperine in a single dose of 80 mg. The plasma level of benproperine was determined by HPLC. Results The in vivo process of benproperine in Beagle dogs fit the one compartment model. The main pharmacokinetic parameters were C.,ax(133.52±58.23) ng/mL; Tm=(2.96±0.58)h; tl/2(2.27±0.51)h; AUC(0-24) (985. 16± 135.23)ng/(h· mL). Conclusion The established HPLC method is simple, rapid and can be suitable to the in vivo concentration determination of benproperine and its pharmacokinetic study.
