PTEN基因mRNA和蛋白表达与胃癌病理生物学行为的关系

摘    要:目的 :观察PTEN基因在胃癌发展不同阶段中的表达 ,探讨其作用机制。方法 :采用RT PCR方法和SABC免疫组化方法 ,对 4 9例胃癌及其相应的癌旁非癌胃粘膜中PTENmRNA和PTEN蛋白表达进行检测。结果 :PTENmRNA在非癌粘膜、早期胃癌、进展期胃癌的阳性表达率分别为 76 .3% ,4 5 .5 % ,34.2 % ;PTEN蛋白的阳性表达率为 10 0 % ,6 3.6 % ,4 7.4 %。进展期胃癌与非癌粘膜比较差别均十分显著 (P <0 .0 1)。两种检测方法具有相关性 (P <0 .0 5 )。PTEN表达下调与胃癌淋巴结转移和临床分期相关 (P <0 .0 5 )。胃癌组织学恶性行为与PTEN表达相关 (P <0 .0 5 )。结论 :PTEN基因可能参与胃癌的侵袭和转移过程。

Abstract:Objective: The aim of this study was to detect the mRNA and protein expression of PTEN gene in gastric carcinoma, and to investigate the significance of PTEN gene expression in progression of gastric carcinoma. Methods: The mRNA and protein expressions of PTEN were evaluated by RT-PCR and immunohistochemistry in 49 cases of gastric carcinoma and the adjacent normal mucosa. Results: The positive rates of PTEN mRNA expression were 76.3%, 45.5%, and 34.2% in adjacent normal mucosa, early and advanced gastric carcinoma, respectively. The positive rates of PTEN protein expression were 100%, 63.6%, and 47.4% in corresponding tissues. And significant difference in mRNA and protein expression of PTEN was also found between advanced gastric carcinomas and the adjacent normal mucosa (P<0.01). The results from RT-PCR and immunohistochemistry were coincident (P<0.05). Loss or reduced expression of PTEN was significantly correlated with tumor clinicopathological stage and lymph node metastasis gastric carcinoma (P<0.05). Conclusion: Decreased expressions of mRNA and protein of PTEN gene were significantly correlated with progression and pathobiological behaviors of the stomach carcinoma.
