Ras、c-myc、p~(53)癌基因蛋白在喉癌及癌旁 不同距离组织中表达的研究

摘    要:研究癌基因及抗癌基因在喉癌及癌旁不同距离处的表达特点及意义。方法:应用LSAB微波 免疫组化法,检测了ras,c- myc及 p53在 30例喉癌、边缘区、癌旁 0.5cm、1.0cm、1.5cm、2.0cm处粘膜和4例正 常喉粘膜中的表达。结果;①ras、c-myc、p53在喉癌组织中有较高的阳性表达率,分别为86.7%(26/30)、96.7% (29/30)、63.3%(19/30),与癌旁及正常组织相比,差异显著(P<0.01),ras、c-myc在正常组织中有低量表达,p53 从癌旁1.0cm处开始出现表达。②两种以上癌基因蛋白产物同时表达阳性者在喉癌组织为28例(93.3%),其中 ras+c-myc+p53者为17例(56.7%)。癌旁组织中也存在着复合表达,在正常组织中无一例联合表达。③ras、c -myc、p53的单独及联合表达依正常粘膜→癌旁2.0cm→1.5cm→1.0cm→0.5cm→边缘区→癌依次增高,多 数在癌旁1.0cm处与0.5cm以内处的表达存在有显著差异(P<0.05)。④三种癌基因产物的表达与喉癌的分化 程度无关(P>0.05)。结论:ras、c-myc、p53基因改变均参与了喉癌?

Abstract:: To study occurrence and significant of oncogene and anti-oncogene expression in laryngeal carcinoma and different mucosa. Methods: With the LSAB-microwave innnUnohistochendcal technique. The expression of ras, c-myc and p53 was studied in different sites: 30 cases of laryngeal squmous cell carcinoma (LSCC), border area, adja cent mucosa which was 0.5cm, 1.0cm, 1.5cm, 2.0 cm away from cancer and four cases normal laryngeal Results: ① The positive rates of ras, c-myc, p53 expression in carcinoma were 86.7%, 96.7%, 63s.3% respectively, which was higher than that in both adjacent mucosa and normal mucosa (P<0.01). The expression of p53 starts at the place 1.0cm away from the tumor. ②The positive rates of co-expression of two or three oncogenes were 93.3% (28/30) in carcinoma. Co-expression was also found.in adjacent mucosa to cancer, no co-expansion was seen in any of the 4 normal cases. ③The positive rates of both single expression and co-expreSsion of p21, p62, and p53 increase as tissue progress in sequence of normal mucosa, 2.0cm, 1.5cm 1.0cm, 0. 5cm distant mucosa adjacent to carcinoma bother area and carcinoma. Significant different expression in most of them can be seen at places 1.5cm and 1.0cm distant form the tumor (P<0s.05). ④There is no correlation between expression of ras, c-myc and p53 and histological grade (P>0.05). Conclusion: The alteration of c-myc, ras and p53 genes are involved in the carcinogenesis of larynx and maybe an early event in developement in LSCC. They may play a combined role in laryngeal carcinoma occuring. The tissue 0.5cm abjacent to the tumor should be regarded as dangerous lesion.
