Rsf-1在胃癌和癌前病变组织中的表达及临床意义

摘    要:目的:染色质重塑因子1(chromatin remodeling factor 1,Rsf-1)可作为肿瘤靶向治疗的靶点,因此探讨其在胃癌(gastric cancer)及癌前病变(precancerous lesions)组织中的表达及临床意义。方法:采用免疫组化SP法检测30例萎缩性胃炎、30例肠上皮化生、22例不典型增生Rsf-1蛋白表达水平。并检测64例胃癌与配对癌旁组织(手术切缘距离肿瘤>5 cm)中Rsf-1和p53蛋白表达水平并分别分析与胃癌临床病理参数间的关系,同时分析Rsf-1与p53在胃癌中表达的相关性。结果:Rsf-1在胃癌、肠上皮化生、不典型增生组织中的阳性表达率均高于癌旁组织(P<0.05)。Rsf-1在胃癌中表达与淋巴结转移有关(P<0.05)。p53在胃癌中的阳性表达率高于癌旁组织,其表达与淋巴结转移有关(P<0.05)。p53和Rsf-1在胃癌中的表达存在正相关性(r=0.38,P<0.05),两者在胃癌中共阳性表达与淋巴结转移有关(P<0.05)。结论:Rsf-1可能参与了胃癌病变的发生及发展,对胃癌早期筛查具有重要指导意义。Rsf-1和p53两者共阳性表达对于预测胃癌淋巴结转移可能具有一定的参考价值。

Abstract:Objective:Chromatin remodeling factor can be used as a target for tumor targeted therapy.Therefore,to explore its the expression and clinical significance in gastric cancer and precancerous lesions.Methods:To explore the protein expression of Rsf-1 in 30 cases of atrophic gastritis,30 cases of intestinal metaplasia mucosa,22 cases of atypical hyperplasia mucosa.The expression levels of Rsf-1 and p53 protein in 64 cases of gastric cancer and their matched adjacent tissues (surgical margin distance>5 cm) were detected,and the relationship between Rsf-1 and p53 protein expression and clinicopathological parameters of gastric cancer was analyzed.Immunohistochemistry was used to detect Rsf-1 and p53 protein expression in different tissues by streptomycin avidin-peroxide polymerase chain connect(SP).Results:The positive expression rate of Rsf-1 in gastric cancer,intestinal metaplasia,atypical hyperplasia was significantly higher than those in adjacent tissue to carcinoma (P<0.05).The expression of Rsf-1 in gastric cancer was associated with lymph node metastasis(P<0.05).The positive expression rate of p53 in gastric cancer tissue was higher than the adjacent tissue to carcinoma,and the expression was related to lymph node metastasis (P<0.05).There was a positive correlation between the expression of p53 and Rsf-1 in gastric cancer (r=0.38,P<0.05),and the positive expression of the two proteins in gastric cancer was related to lymph node metastasis (P<0.05).Conclusion:Rsf-1 may be involved in the occurrence and development of gastric cancer pathological changes,and it has important guiding significance for early screening of gastric cancer.The co-expression of Rsf-1 and p53 for predicting lymph node metastasis of gastric cancer may have a certain reference value.
