纳洛酮对大鼠脑出血Caspase-3表达的影响

目的：探讨纳洛酮（NAL）治疗对大鼠急性脑出血（ICH）模型Caspase-3表达的影响。方法：健康雄性SD大鼠12只,体重260~310 g,随机分为NAL治疗组、对照组各6只。于大鼠尾状核区注入自体不凝血制备ICH模型,治疗组予以NAL保护,对照组予以等量生理盐水。术后72h将血肿周围部位脑组织取材、固定、染色,观察脑组织Caspase-3表达情况。结果：经NAL保护的动物血肿周围脑组织内出现的Caspase-3阳性细胞数量较少,与对照组的差异具有统计学意义（P〈0.05）。结论：对大鼠急性ICH模型进行NAL保护可以减少凋亡促进物质Caspase-3的表达。

Effects of NAL on the Expression of Caspase-3 in Rat Model of Acute ICH

Objective：To investigate the effect of NAL on the expression of Caspase-3 in rat model of acute ICH.Methods：Twelve adult healthy male Sprague Dawley rats of 260～310g in weight were divided randomly into two equal groups,namely NAL group and control group.An ICH model was established by injecting uncoagulated autologous blood into caudate nucleus.Then NAL was administered to the NAL group while the same volume of physiological brine was administered to the control group.Seventy-two hours later after the operation,the brains adjacent to the hemerrhage were removed and fixed and Caspase-3 were stained.The numbers of positive cells of Caspase-3 staining were counted then.Results： In NAL group,the numbers of Caspase-3-positive cells were significantly smaller than those in control group（P〈0.05）.Conclusion：NAL can significantly reduce the expression of Caspase-3,a proapoptotic factor,in rat model of ICH.
