低氧预适应对大鼠脑缺血再灌注神经细胞凋亡及Bcl-2、Bax表达的影响

摘    要:目的 :探讨低氧预适应对大鼠大脑中动脉 (MCA)缺血再灌注神经细胞凋亡 ,Bcl 2、Bax的表达及行为学计分的影响。方法 :成年Wistar大鼠 75只 ,随机分为 3组 :假手术组 ,缺血再灌注组 (对照组 )和低氧预适应组。采用动脉线栓法制作大鼠脑缺血再灌注模型 ,假手术组除不插线外 ,其余同缺血再灌注组 ,低氧预适应组在低氧预适应后 12h制作脑缺血再灌注模型 ,低氧预适应后在大鼠MCA缺血 3h再灌注 1h、6h、12h、2 4h、4 8h不同时间点 ,进行脑切片的TUNEL染色 ,Bcl 2、Bax的免疫组化检测及大鼠的行为学计分。结果 :低氧预适应大鼠的神经细胞凋亡、Bax的表达和行为学计分均较对照大鼠减少 ,Bcl 2的表达较对照组增高 ,差异均有统计学意义 (P <0 .0 5 )。结论 :低氧预适应的脑保护及改善神经功能的作用与其抑制神经细胞的凋亡有关 ;而增加Bcl 2及减少Bax的表达 ,可能是实现保护作用、改善大鼠神经功能的机制之一。

Abstract:Aim: To study effects of hypoxic preconditioning on the apoptosis of neural cells and the expression of Bcl-2, Bax in rat cerebral ischemia-reperfusion. Methods: A total of 75 adult male Wistar rats were randomly divided into sham operation group, ischemia-reperfusion group, and hypoxic preconditioning group. The model was established by inserting the thread to MCAO, and preconditioning group was performed 12 hours before ischemic by placeing rats in the hypoxic chamber of 8% oxygen for 3 hours. The sham operation group was performed by the same way except for inserting the thread.After 1 h, 6 h, 12 h, 24 h,and 48 h of cerebral ischemia for 3 hours and reperfusion, the rat brain sections were applied for TUNEL staining, Bcl-2 and Bax with immunohistochemistry staining. Meanwhile, the behavioral scores of rats were evaluated. Results: The apoptosis of neural cells, the expression of Bax and the behavioral scores in the hypoxic preconditioning group significantly decreased as compared with the control group. While the neurological function was significantly improved. However, the expression of Bcl-2 of the hypoxic preconditioning group significantly increased as compared with the control group(P<0.05). Conclusion: The mechanism of brain protection and neurological function recovery with hypoxic preconditioning may relate to inhibit the apoptosis of neural cells, increase the expression of Bcl-2, and decrease the expression of Bax.
