摘    要:目的:建立用高效液相色谱同时测定何首乌中已知有效成分二苯乙烯苷、大黄素、大黄酸、大黄酚、大黄素甲醚和芦荟大黄素含量的方法。方法:采用高效液相色谱法,Agilent TC-C18色谱柱(4.6mm×250mm,5μm),甲醇-0.1%磷酸溶液梯度洗脱,1.0mL/min,254nm,30℃。结果:6种化合物呈现较好的回归方程和相关系数,相关系数达到0.9999。二苯乙烯苷、大黄素和大黄素甲醚的平均回收率分别为97.39%、98.65%和98.35%。结论:该方法快速准确,精密度高,稳定性好,大大缩短了测定时间,对现行药典方法有了很大的改进,为何首乌的质量控制与含量测定提供了重要的参考。

Abstract:Objective: To establish a method for determination of stilbeneglycoside and anthraquinone in Polygonum multiflorum Thunb by HPLC stimultaneously.Methods: The separation was performed on Agilent TC-C18(4.6mm×250mm,5μm) column with method-0.1% phosphoric acid water solution gradient elution.The flow rate was 1.0mL/min,the wavelength of UV detector was set at 254nm and the column temperature was set at 30℃.Results: The sample size of stilbeneglycoside and anthraquinone showed respectively a good linear relationship(r=0.9999).The average recovery of stilbeneglycoside was 97.39%;the average recovery of emodin was 98.65%;the average recovery of physcion was 98.35%.Conclusion: The method is simple,accurate and with good reproducibility.
