慢性阻塞性肺病患者外周血中0D34^+细胞数量变化

摘    要:目的:观察中、重度慢性阻塞性肺病(COPD)患者外周血中CD34^+细胞数量变化。方法:按入选标准收集25例中、重度COPD患者外周血,应用密度梯度离心法分离单个核细胞,采用CD34^+正分选试剂盒进行磁珠纯化,用流式细胞分析仪分析其纯度,同时对入选患者行常规心电图、肺功能、心动超声及血气检查。同期16例性别、年龄无差异志愿者作为对照组,入选标准:年龄〈70岁;肺功能正常;没有临床症状;无吸烟史。结果:经过分离纯化可得到93%表达CD34^+细胞。COPD患者组与对照组外周血中的CD34^+细胞数分别是(19.99±11.11)×10^4、(32.75±18.25)×10^4,患者组CD34^+细胞数明显少于对照组(P〈0.05),CD34^+细胞数与患者病程呈负相关(r=-0.689,P〈0.05)。结论:COPD患者外周血中CD34^+细胞数降低,与患者病程呈负相关。

Change of EPCs in perpherail blood of COPD patients

Abstract:Objective. To observe the change of EPCs in perpherail blood of chronic obstructive pulmonary disease (COPD) patients. Methods. Based on the selection standard, peripheral blood was col- lected from media and severe COPD patients. Peripheral blood mononuclear cells (PBMCs) were obtained by standard Ficoll gradient centrifugation, and purified by CD34^+ positive selection cocktail. All subjects received a complete clinical assessment including pulmonary function tests, a 12-lead ECG and blood gas analysis. The selection standard of control group was as follows, age〈70 years old, normal pulmonary function, no clinical symptoms or smoking history. Results: 93% CD34^+ cells was obtained after isolation and purification. The count of CD34^+ cells was (19.99±11.11) ×10^4 and (32.75±18.25)×10^4in patients group and control group, respectively. The count of patients group was significantly lower (P〈0.05). There were negative correlation between CD34^+ cell counts and course of COPD(r=-0. 689 ,P〈0.05). Conclusions: The count of CD34^+ is decreased, and is negatively correlated with course of COPD.
