高压氧对大鼠脑缺血再灌注后神经细胞凋亡的影响

摘    要:目的 探讨高压氧对大鼠脑缺血再灌注后神经细胞凋亡的影响.方法 成年健康雄性Wistar大鼠32只,分为模型对照组、高压氧暴露组,2组再各分4小组,分别为脑缺血1 h冉灌注后12、24 h,3、5 d组,每组4只.应用线栓法制作大鼠左侧大脑中动脉阻塞再灌注模型(MCAO),并进行高压氧暴露,应用HE染色和细胞凋亡检测,观察神经细胞病理变化和细胞凋亡分布.结果 脑缺血再灌注后12 h即出现神经细胞凋亡表达,随着时间的延长凋亡细胞的数量逐渐增加,至24 h达高峰,以后逐渐减弱,至5 d仍有表达;高压氧暴露组凋亡细胞的数量相对较少,其变化规律与对照组相似,但是同一时间点比较均明显低于对照组.结论 高压氧可以抑制脑神经元细胞凋亡,保护脑神经细胞,有利于促进神经功能恢复.

Abstract:Objective To study effects of hyperbaric oxygen (HBO) on apoptosis of nerve cells following focal cerebral isehemia reperfusion in rats. Methods The rat middle cerebral artery occlusion reperfusion (MCAO) model was developed by using the intraluminal filament method. 32 rats were randomly divided into2 groups: the control group (n=16) and the HBO group (n=16). Then, the 2 groups, each consisting of 4 animals, were further subdivided into 4 groups, i.e. the hour 12 reperfusion group, the hour 24 reperfusion group, the day 3 reperfusion group and the day 5 repcrfusion group. Following 1-hour cerebral ischemia, the experimental animals were exposed to HBO. Then, the expression of apoptosis was determined with HE staining and cell apoptosis detection. Pathological changes in neural cells and distribution of apoptosis were observed in the study. Results In the control group, the apoptosis of neural cells could be detected 12 hours after reperfusion following cerebral ischemia, the number of apoptosis cells gradually increased with the extension of time, reached a maximum at hour 24, then subsided gradually. In the HBO group, the number of cells was significantly lower than those in the control group at the same time point. Conclusions HBO could inhibit the apoptosis of neurons, protect neural cells and help to enhance recovery of neural functions following cerebral ischemia.
