摘    要:目的 :探讨玄参提取物 (SnPs)对局灶性缺血后脑组织的保护作用及对脑血流量的影响。方法 :雄性SD大鼠 (2 4 2±s 7)g 4 0只随机分 5组 (均n =8) :假手术组、模型对照组、3个不同剂量SnPs治疗组 (1,5 ,10mg·kg- 1尾静脉注射 )。大脑中动脉线栓法建立局灶性脑缺血模型 (MCAO) ,缺血 2 4h后 ,用神经功能评分和梗死体积改变来评价SnPs对脑缺血的治疗作用。另将MCAO模型缺血SD大鼠分成 4组 (模型对照组n =8,3个不同剂量 1,5 ,10mg·kg- 1SnPs治疗组均n =6 )用于测脑血流量 ,用激光多普勒仪分别在 6个时间点 (缺血前 ,缺血即刻 ,缺血后 30min和 2h ,以及缺血 2h再灌 30min ,2h)记录双侧的皮层血流量。结果 :脑缺血 2 4h后 ,各SnPs治疗组与模型对照组相比 ,脑梗死体积明显减少 (P <0 .0 5 ) ,神经功能均明显改善 (P <0 .0 1) ;但在此剂量范围内无明显的剂量依赖性。各SnPs治疗组与模型组相比 ,缺血 2h后皮层CBF明显改善 (P <0 .0 5 ) ,其中 5mg·kg- 1剂量组对于缺血各时间点的血流改善均有显著作用 (P <0 .0 5 )。结论 :SnPs对于脑缺血损伤具有保护作用 ,此作用可能与提高脑血流量有关。

Abstract:AIM: To investigate the protectiveeffect of Scrophularia ningpoensis extracts (SnPs)on focal cerebral ischemic and the influence on cerebral blood flow (CBF). METHODS: MaleSprague-Dawley rats were subjected to middle cerebral artery occlusion (MCAO). Forty rats were randomly divided into five groups (n = 8), sham operation group, model group and SnPs-treatment groupsscores were determined 24 h after MCAO (grading of0 to 4). Ischemic lesion volume was measured by 2,3,5 -triphenyltetrazolium chloride (TTC) staining.CBF was monitored at six time points: beforeMCAO; instant, 0.5 h, 2 h after MCAO and 0.5 h,2 h after reperfusion performed (2 h after MCAO) incontrol group ( received saline). RESULTS: TheSnPs-treated groups showed significant improvementin neurological deficits compared with control group(P<0.01). Lesion volume was (15 ± s 4) % , (14±5) %, (15±3) % and (22±4) % in three treated groups and control groups, respectively. The lesion volume was significantly decreased 24 h afterMCAO in SnPs groups compared with the controlgroup (P < 0.05). In comparison to CBF decrease incontrol group, CBF was significantly improved in theIn other two treated groups, CBF was obviously improved only at the time point of 2 h after MCAO (P< 0.05). CONCLUSION: SnPs has protectiveeffect on cerebral ischemia injury and this effect maybe associated with enhancement of CBF recovery.
