1,25二羟维生素D_3对卵巢癌细胞株HO8910增殖及Skp_2/p27蛋白表达的影响

目的:观察1,25二羟维生素D3[1,25(OH)2D3]对人卵巢癌细胞株HO8910增殖的影响,并且检测其作用下S期激酶相关蛋白(Skp2)、抑癌基因p27的蛋白表达情况,以探讨其作用机制。方法:选用人卵巢癌细胞株HO8910进行体外培养,用MTT比色法测定不同浓度D3[1,25(OH)2D3]对细胞株作用不同时间后细胞生长抑制率,流式细胞仪进行细胞周期分析,免疫细胞化学染色检测Skp2及p27蛋白表达情况。结果:1,25(OH)2D3在160×10-9mol/L以上时,对细胞有抑制作用(P<0.05),且呈浓度时间依赖性;1,25(OH)2D3能增加G0/G1期细胞比例,降低S、G2/M期细胞比例,从而降低细胞增殖指数(P<0.01);能降低Skp2表达,增加p27蛋白表达(P<0.01)。结论:1,25(OH)2D3对人卵巢癌细胞株HO8910有抑制作用,可能通过下调Skp2表达、上调p27蛋白表达,从而抑制癌细胞增殖。

The effect of 1,25(OH)2D3 on growth and expression of Skp2/p27 protein in ovarian cancer cell line HO8910

Objective:To study the effect of 1,25(OH)2D3 on growth in human ovarian cancer cell line HO8910 and its mechanism by detecting the expression of S-phase kinase-interacting protein-2(Skp2) and p27 protein.Methods:HO8910 human ovarian cancer cell line were cultured in vitro and treaed with different concentrations of 1,25(OH)2D3 for 1,2 and 3 days.Cell proliferation was evaluated by MTT assay,distribution of cell cycle were determined by flow cytometry,and the expression of Skp2 and p27 protein were detected by using immunocytochemistry.Results:1,25(OH)2D3 inhibited the growth of HO8910 cells at concentrations of 160×10-9 mol/L or more(P<0.05).It assumed concentration and time dependent effect.1,25(OH)2D3 increased the rates of G0/G1,decrease the rates of S and G2/M,and so decreased cells PI(P<0.01).It decreased the espression of Skp2 and increased p27 protein(P<0.01).Conclusion:1,25(OH)2D3 can inhibit the growth of HO8910 cell line.It may inhibit cancer cell proliferation through down-regulation of Skp2 and up-regulation of p27 protein.
